摘  要:目的观察人参皂苷Rb3通过Neuregulin-1/ErbB信号通路对心肌梗死大鼠VE-cadherin,NRG-1,Erb B2,Erb B4表达的影响。方法 60只大鼠随机分为Sham组、Model组、G-Rb3-L组(10 mg/kg)、G-Rb3-M组(20 mg/kg)、G-Rb3-H组(40 mg/kg)及阳性药物组(地尔硫卓20 mg/kg),每组各10只。造模后,G-Rb3-L组、G-Rb3-M组、G-Rb3-H组及阳性药物组给予相对的药物,Model组和Sham组大鼠给予等体积的生理盐水。HE、Masson染色分别观察心肌组织的病理变化与心肌纤维化程度;ELISA法检测血浆中的VE-cadherin的水平;Western blotting法检测心肌组织NRG-1、ErbB2和ErbB4蛋白的表达。结果与Sham组比较,Model组大鼠的心肌细胞炎症浸润及心肌纤维化程度严重,心肌梗死面积、VE-cadherin水平上升(P<0.05),NRG-1,Erb B2和Erb B4蛋白表达减少(P<0.05);与Model组比较,人参皂苷Rb3干预的大鼠的心肌细胞炎症浸润及心肌纤维化程度减轻,心肌梗死面积、VE-cadherin表达水平降低(P<0.05),NRG-1,Erb B2和Erb B4蛋白表达增加(P<0.05)。结论人参皂苷Rb3可通过调控Neuregulin-1/ErbB信号通路降低心肌梗死大鼠的心肌细胞的炎症及心肌纤维化程度,减少心肌梗死面积,减轻心肌损伤。Objective To observe the effect of ginsenoside Rb3 on the expression of VE-cadherin,NRG-1,ErbB2 and ErbB4 in rats with myocardial infarction by Neuregulin-1/ErbB signaling pathway.Methods 60 rats were randomly divided into Sham group,Model group,G-Rb3-L group(10 mg/kg),G-Rb3-M group(20 mg/kg),G-Rb3-H group(40 mg/kg)and positive drug group(Diltiazem 20mg/kg),with 10 rats in each group.After modeling,the G-Rb3-L group,the G-Rb3-M group,the G-Rb3-H group and the positive drug group were given relative drugs,and the Model group and the Sham group were given an equal volume of physiological saline.The pathological changes of myocardial tissue and the degree of myocardial fibrosis were observed by HE and Masson staining.The levels of VE-cadherin in plasma were detected by ELISA.The expressions of NRG-1,ErbB2 and ErbB4 in myocardial tissue were detected by Western blotting.Results Compared with the Sham group,the myocardial infiltration and myocardial fibrosis were severe in the model group,myocardial infarct size,VE-cadherin protein were significantly increased(P<0.05),and the expression levels of NRG-1,ErbB2,and ErbB4 proteins were significantly reduced(P<0.05).Compared with the Model group,after intervention with ginsenoside Rb3,the cardiomyocyte inflammatory infiltration and myocardial fibrosis were reduced,myocardial infarct size and VEcadherin protein expression were significantly reduced(P<0.05).NRG-1,ErbB2 and ErbB4 protein expression was significantly increased(P<0.05).Conclusion Ginsenoside Rb3 through Neuregulin-1/ErbB signaling pathway can effectively reduce the myocardial inflammation and myocardial fibrosis in myocardial infarction rats,and reduce myocardial infarct size and myocardial injury.
