摘 要:为了探索热应激降低小鼠早期囊胚后续发育能力的作用机理,试验采用形态学分析的方法,将试验动物昆明种小鼠分为39℃热应激组、41℃热应激组和对照组(37℃),从亚细胞水平观察体外热应激处理后小鼠早期囊胚细胞骨架的变化。结果表明:对照组和39℃热应激组小鼠早期囊胚紧密连接,桥粒未发生明显变化;而41℃热应激组小鼠早期囊胚桥粒连接结构不完整,两侧附着微丝减少,可见到明显的缝隙,卵周隙明显增大,微绒毛减少、弯曲,且细胞器的修复能力变差。说明细胞骨架的破坏是热应激降低小鼠早期囊胚后续发育能力的作用机理之一。In order to explore the mechanism of thermal stress to reduce the ability of early blastocysts development in mice, morphological analysis was used in the experiment to determine ultrastructural changes of cytoskeleton of mice early blastocysts. The mice were randomly divided into heat stress group at 39℃,heat stress group at 41℃, and control group(37℃). Changs of early blastocysts cytoskeleton in mice ofter heat stress in vitro observed from subcellular level. The results showed that the early blastocysts in the control group and the heat stress group at 39℃. were closely connected, and there was no significant change in the desmosome. However, in the heat stress group at 41℃, the connection of desmosomes was disrupted, the microfilament of desmosomes has decreased, the gap became more distinc.The space between the nuclear envelope and the perivitelline membrane enlarged. The microvillus reduced and bended. The ability of organelles to repair deteriorated indicates the injury of the cytoskeleton may be one of the mechanism of thermal stress to reduce the ability of early blastocysts.
