摘 要:目的观察败酱总黄酮对局灶性脑缺血再灌注大鼠的神经保护作用及炎症因子的影响。方法将98只Wistar大鼠随机分为假手术组,模型组,尼莫地平组(20 mg·kg^(-1)),脑络通组(500 mg·kg^(-1)),败酱总黄酮高、中、低剂量组(200、100、50 mg·kg^(-1))。灌胃体积为20 mL·kg^(-1),每天给药1次,连续给药7 d。末次给药后采用线栓法建立大鼠局灶性脑缺血再灌注模型。采用ELISA法检测脑组织匀浆中白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、肿瘤坏死因子α(TNF-α)、B细胞淋巴瘤因子2(Bcl-2)、Bcl-2相关X蛋白(Bax)、半胱氨酸蛋白酶-3(Caspase-3)、细胞间黏附分子1(ICAM-1)水平;氯化三苯基四氮唑(TTC)染色观察并测定脑梗死面积百分比;免疫组织化学法检测脑组织中神经生长因子(NGF)、核因子(NF)-κBp65表达情况。结果与假手术组比较,模型组大鼠脑组织脑梗死面积百分比、ICAM-1、IL-6、IL-1β、TNF-α、Bax、Bcl-2、Caspase-3水平以及NGF、NF-κBp65免疫阳性表达水平显著升高(P <0.01)。与模型组比较,败酱总黄酮各剂量组的大鼠脑组织ICAM-1、IL-6、IL-1β、Bax、Caspase-3含量水平均显著降低(P <0.05,P <0.01),Bcl-2含量水平及NGF免疫阳性表达水平均显著升高(P <0.01);败酱总黄酮高、中剂量组的大鼠脑梗死面积百分比、TNF-α含量水平及NF-κBp65免疫阳性表达水平均明显降低(P <0.05,P <0.01)。结论败酱总黄酮对局灶性脑缺血再灌注损伤有较好的保护作用,可以减少脑梗死面积,保护脑组织免受炎症因子的损伤,增加神经细胞营养因子保护神经元,减少神经细胞的凋亡,激活脑细胞自我保护,减少脑缺血再灌注损伤。Objective To observe the neuroprotective effect of total flavonoids from Patrinia on focal cerebral ischemia-reperfusion rats and the effect on inflammatory factors. Methods Ninety-eight Wistar rats were randomly divided into Sham operation group,model group,nimodipine group(20 mg·kg^-1),Naoluotong group(500 mg·kg^-1)and high,medium and low dose groups(200,100 and 50 mg·kg^-1) of Patrinia total flavonoids. The volume of gastric administration was 20 mL·kg^-1 once a day for 7 d. The rat model of focal cerebral ischemia-reperfusion was established after the last administration by the method of thread bolt. ELISA was used to detect the levels of interleukin-6(IL-6),interleukin-1β(IL-1β),tumor necrosis factor α(TNF-α),B cell lymphoma factor 2(Bcl-2),Bcl-2 related X protein(Bax),cysteine protease-3(Caspase-3) and intercellular adhesion molecule 1(ICAM-1) in brain homogenate. TTC staining was used to observe and measure the percentage of cerebral infarction area. Immunohistochemistry was used to detect the expression of nerve growth factor(NGF) and nuclear factor(NF)-κBp65 in brain tissue. Results Compared with the Sham operation group, the cerebral infarction area percentage, ICAM-1, IL-6, IL-1β, TNF-α, Bax, Bcl-2, Caspase-3 levels and NGF, NF-κBp65 immunoreactive levels in the model group were significantly increased(P < 0.01). Compared with the model group,the levels of ICAM-1,IL-6,IL-1β,Bax,Caspase-3 in the brain tissue of rats in all dosage groups of Patrinia total flavonoids decreased significantly(P < 0.05,P < 0.01),and the levels of Bcl-2 and NGF immunoreactive expression increased significantly(P < 0.01). The percentage of cerebral infarction area,the level of TNF-α and the level of NFsignificantly reduced(P < 0.05,P < 0.01). Conclusion Patrinia flavonoids have a good protective effect on focal cerebral ischemia-reperfusion injury,which can reduce the area of cerebral infarction,protect brain tissue from inflammatory factors,and increase nerve cell nutrition factors to protect neurons,reduce apop
