摘 要:目的观察复方当归提取物(当归、三七、陈皮等组成)对高脂高糖饮酒致高脂血症模型大鼠的影响。方法采用高脂高糖饲料喂养复合梯度饮酒制备高脂血症模型大鼠,造模成功后灌胃给予复方当归提取物连续10周。给药期间,测定大鼠抓力、痛阈、眩晕时间和自主活动次数;检测血清中胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-c)和低密度脂蛋白胆固醇(LDL-c)水平;末次给药后,测定血清载脂蛋白A1(APO-A1)、胆固醇酯转移蛋白(CETP)和卵磷脂胆固醇酰基转移酶(LCAT)水平;HE染色观察大鼠肝脏和胸主动脉组织形态学,Masson染色观察胸主动脉和肝脏纤维化情况;免疫组化法检测胸主动脉中细胞间粘附分子-1(ICAM-1)表达。结果复方当归提取物能降低模型大鼠眩晕时间(P <0.01),提高痛阈值和自主活动次数(P <0.01,P <0.05);降低血清TC、TG、LDL-c水平(P <0.01,P <0.05),升高血清HDL-c、APOA1、LCAT水平(P <0.01,P <0.05);抑制胸主动脉中膜增厚及内皮细胞脱落,并改善胸主动脉和肝脏中胶原沉积;抑制胸主动脉中ICAM-1的表达。结论复方当归提取物可能通过升高LCAT、APO-A1水平和抑制ICAM-1表达,从而改善高脂高糖饮酒致大鼠血脂异常、肝脏和胸主动脉病变。Objective To observe the effect of compound Angelica sinensis extract on hyperlipidemia rats induced by high fat and high glucose compound alcohol. Methods The rats were fed with high fat and high glucose compound alcohol diet to prepare the hyperlipidemia models, and the compound Angelica sinensis extract was given for 10 weeks after the models were successful established. During the administration,the grasping force,pain threshold,vertigo time and autonomic activity of rats were observed and measured. The serum levels of Total cholesterol(TC),Triglyceride(TG),High density lipoprotein cholesterol(HDL-c),Low density lipoprotein cholesterol(LDL-c)were measured. After the last administration, Apolipoprotein A1(APO-A1), Cholesterol ester transfer protein(CETP),Lecithin cholesterol acyltransferase(LCAT) were measured,and the liver and thoracic aorta tissue of rats was observed by HE. Morphology and Masson staining were used to observe the fibrosis of the liver and thoracic aorta. Immunohistochemical staining was used to detect the expression of intercellular cell adhesion molecule-1(ICAM-1) in the thoracic aorta. Results Compound Angelica sinensis extract can reduce the vertigo time(P <0.01),increase the pain threshold and the number of spontaneous activities(P < 0.01,P < 0.05),reduce the levels of serum TC,TG and LDL-c(P < 0.01,P < 0.05),and increase the contents of HDL-c,APO-A1 and LCAT(P < 0.01,P < 0.05),inhibit the thickening of the thoracic aorta and endothelial cell shedding,and improves the deposition of collagen fibers in the thoracic aorta and liver,inhibit the expression of ICAM-1 in the thoracic aorta. Conclusion Compound Angelica sinensis extract may improve the dyslipidemia, liver and thoracic aortic lesions of hyperlipidemia rats by increasing the levels of LCAT,APO-A1 and inhibiting the expression of ICAM-1.
