摘 要:目的观察菊花总黄酮含药血浆对大鼠泪腺上皮细胞干眼症模型中Bax mRNA及Bcl-2 mRNA表达的影响。方法将培养生长状态良好的大鼠泪腺上皮细胞随机分为无雄激素培养组(空白组)、雄激素培养组(雄激素组)、无雄激素培养黄酮组(菊花总黄酮组),分别加入空白血浆、丙酸睾酮、菊花总黄酮含药血浆干预48 h,然后加入100μmol/L H_2O_2继续培养60 min诱导细胞凋亡。RT-PCR法检测泪腺上皮细胞中Bax mRNA及Bcl-2 mRNA的表达情况。结果雄激素组和菊花总黄酮组Bax mRNA表达量均明显低于空白组(P均<0.05),Bcl-2 mRNA表达量均明显高于空白组(P均<0.05),雄激素组和菊花总黄酮组比较差异均无统计学意义(P均>0.05)。结论菊花总黄酮含药血浆有可能通过抑制泪腺上皮细胞中Bax mRNA表达及促进Bcl-2 mRNA表达的途径,起到抑制泪腺上皮细胞凋亡的作用。Objective It is to observe the effects of Chrysanthemum total flavonoid-containing plasma on the expression of BaxmRNA and Bcl-2 mRNA in lacrimal gland epithelial cells of rat model of xerophthalmia. Methods The rat lacrimal gland epithelial cells with good growth state were randomly divided into the no-androgen-free culture group(blank group), the androgen culture group(androgen group) and the no-androgen cultured flavonoid group(chrysanthemum total flavonoid group) which were treated with blank plasma, testosterone propionate, chrysanthemum total flavonoids containing plasma for 48 h, then 100 μmol/L H2O2 was added for 60 min to induce apoptosis. The expression of Bax mRNA and Bcl-2 mRNA in lacrimal gland epithelial cells was detected by RT-PCR. Results The expression of Bax mRNA in androgen group and total flavonoids of chrysanthemum group was significantly lower while the expression of Bcl-2 mRNA was significantly higher than that in the blank group(P<0.05). There were no significant differences in these indexes between androgen group and chrysanthemum total flavonoids group(P>0.05). Conclusion The total flavonoid-containing plasma of chrysanthemum may inhibit the apoptosis of lacrimal gland epithelial cells by inhibiting the expression of Bax mRNA and promoting the expression of Bcl-2 mRNA in lacrimal gland epithelial cells.
