摘 要:目的研究西南地区人乳头瘤病毒6型(HPV-6)和11型(HPV-11)E6/E7基因多态性,为低危型HPV的防治提供分子水平。方法对HPV-6和HPV-11阳性标本扩增其目的基因,测序并对比Genenbank参考序列,对变异初步分析;PAML 4.8软件作用于E6/E7基因的选择压力。结果 HPV-6E6/E7基因共12个突变位点,其中有3个是首次发现的突变位点,5/12为非同义突变;HPV-11E6/E7中,共计5个单核苷酸突变,具有2/5非同义和3/5同义突变。选择压力结果显示,只有HPV-6E7基因有3个正向选择位点,分别是4R**,34E**和52F**。结论 HPV-6E6和E7序列在分析的群组中高度保守,表示HPV-6在进化的过程中,形成了有利于生存的分离株。HPV-11突变未发现正向选择位点,表明选择的变异对HPV-11来说适应环境较困难。Objective To investigate the polymorphism of E6/E7 genes of human papillomavirus type 6 (HPV-6) and human papillomavirus type 11 (HPV-11) in southwest China and provide the molecular level for the prevention and treatment of low-risk HPV. Methods The target genes from HPV-6 and HPV-11 positive samples was amplified, sequenced and compared with the Genenbank reference sequences for preliminary analysis of mutation. The software PAML 4.8 was used for the selection pressure of E6/E7 gene. Results There were 12 mutation sites in the HPV-6 E6/E7 gene, of which 3 were the mutation sites found for the first time and 5 were non-synonymous mutations. There were 5 single nucleotide mutations with 2 nonsynonymous mutations and 3 synonymous mutations in the HPV-11 E6/E7 gene. The results of selection pressure showed that the HPV-6 E7 gene had three positive selection sites which were 4R^**, 34E^** and 52F^**. Conclusion The HPV-6 E6/E7 sequences are highly conserved in the analyzed cohort, which indicates that HPV-6 forms survival-friendly isolates during the evolution process. No positive selection sites are found in the HPV-11 mutation, which indicates that the selected mutation is difficult to adapt to the environment for HPV-11.
