摘  要:目的:探讨补阳还五汤对创伤性脑损伤小鼠皮质运动区神经元的保护作用及可能机制。方法:将144只小鼠随机分为假手术组、模型组和用药组,并按给药时间分为1、3、7、14天四个亚组,造模后,用药组给予补阳还五汤治疗,假手术和模型组给予生理盐水。观察记录小鼠行为学变化;尼氏染色法,观察损伤周围区神经元的形态学改变;免疫组织化学染色法,检测损伤周边区Bcl-2、Bax和Caspase-3蛋白的阳性表达情况。结果:与模型组相比,用药组小鼠随着给药时间增加,运动行为表现改善明显,具有显著统计学意义(P<0.01);尼氏染色显示,用药组脑损伤区域呈现正常形态神经元增多,异常凋亡细胞减少的趋势;免疫组化定量分析结果显示,用药各组小鼠脑损伤周围区域Bcl-2蛋白的表达明显增强,而Bax和Caspase-3蛋白表达则减弱,两组间各时间点差异大部分具有统计学意义(P<0.01或P<0.05)。结论:补阳还五汤对创伤性脑损伤小鼠皮质运动区神经元有较为明显的保护作用,其可能机制之一为抑制细胞凋亡。Objective:To investigate the neuroprotective effect of Buyang Huanwu Tang(BHT)on lhe cortical motor neurons in the traumatic-train injury mice and the possible mochanism.Methods:144 mice were randomly divided into sham operation group,model group and BHT group*and each group was divided into 4 subgroups based on the administration time points of ld,3d,7d and 14d post-trauma.After the establishment of model.lhe mice of BHT group were administered with medicine,the sham operation and model groups with saline.After observing and recording the mice's alterations in motor behavior佱made their brains slices and treated some with Nissl's staining for observing the pathological changes.Other slices were treated with immunohistochemical staining for detecting the positive expression of Bcl-2.Bax and Caspase-3 proteins in the neurons surrounding the traumatic area and calculating lhe average optical density value(AODV).Results:compared with Model group,the mice's motor function in BHT group was significantly improved and more normal and less apoptotic cells were found near the traumatic area with administration time in creasing.The quantitative analysis of immunohistochemistry revealed the enhanced level of Bcl-2 and reduced level of Bax and Caspase-3 in the contiguity of traumatic area for BHT group compared with Model group,the difference was statistically significant at most time points.Conclusion:BHT may significantly protect the cortical motor neurons in the traumatic-train-injury mice and one possible mechanism is inhibiting the apoptosis of the relevant neuro ns.
