摘  要:目的:探讨TGF-β1诱导的足细胞损伤中miR-155的表达变化及其与足细胞损伤的关系。方法:体外培养小鼠肾小球足细胞,用TGF-β1刺激足细胞构建足细胞损伤模型,用CCK8法检测各浓度梯度(0、4、8、12 ng/ml)、时间梯度(0、24、48、72 h)的细胞增殖活性。实时荧光定量PCR技术检测足细胞损伤模型中synaptpotodin、CD2AP mRNA及miR-155的表达。Western blot法检测足细胞损伤模型中synaptpotodin、CD2AP蛋白的表达。结果:一定浓度的TGF-β1可抑制足细胞增殖,降低细胞存活率(P <0. 05)。TGF-β1诱导的足细胞损伤模型中miR-155表达水平上调(P <0. 05),TGF-β1诱导足细胞损伤后,可引起synaptpotodin、CD2AP mRNA和蛋白表达水平下调,且与miR-155表达水平呈负相关(P <0. 05)。结论:miR-155表达上调与TGF-β1诱导的足细胞损伤有关,可能作为足细胞损伤诊断和治疗的靶点。Objective: To investigate the expression of miR-155 in podocyte injury induced by TGF-β1 and its relation with podocyte injury.Methods: Establishment of podocyte injury model was induced by TGF-β1 in vitro.CCK8 was used to detect the proliferation of cell treated with different TGF-β1 concentration(0,4,8,12 ng/ml) and time(0,24,48,72 h) gradient.The expression of synaptpotodin,CD2 AP mRNA and miR-155 levels in podocyte injury model were detected by qRT-PCR.The expression of synaptpotodin and CD2 AP protein levels in podocyte injury model were detected by Western blot.Results: A certain concentration of TGF-β1 could decrease cell survival rate,thus suppress the cell proliferation(P < 0.05).The expression of miR-155 was increased in podocyte injury model induced by TGF-β1(P < 0.05).The TGF-β1 induced podocyte injury showed decreased expression of synaptopodin and CD2 AP at mRNA and protein levels,and were negatively correlated with the expression level of miR-155(P < 0.05).Conclusion: Upregulation of TGF-β1 is associated with podocyte injury induced by TGF-β1,and may be used as a potential detection and therapeutic target for podocyte injury.
