摘 要:目的:探讨miR-122的表达在胶质瘤中的诊断作用及其与VEGF表达相关性。方法:收集74例胶质瘤患者和74例健康人抗凝标本和血清标本。抗凝标本进行qRT-PCR检测血浆miR-122的含量,应用ROC曲线评价miR-122在胶质瘤中的诊断性能,并与胶质瘤的临床病理特征进行相关分析。运用ELISA定量分析胶质瘤患者及健康人血清VEGF蛋白水平,并进行统计学分析。结果:与健康人相比,胶质瘤患者血浆中miR-122的表达显著降低。ROC分析显示miR-122用于诊断胶质瘤时敏感度和特异度高(灵敏度91. 9%,特异度81. 1%,曲线下面积0. 939)。胶质瘤患者miR-122的表达随着胶质瘤的进展而降低,与WHO分级显著相关。VEGF表达随着胶质瘤分级的增加而升高,与健康对照组相比,Ⅲ-Ⅳ级别升高具有显著性。miR-122和VEGF在胶质瘤中的表达呈负相关。结论:miR-122在胶质瘤患者血浆中呈异常低表达,可作为诊断和预后胶质瘤的生物标志物; miR-122表达的降低,VEGF的升高,为胶质瘤的发生发展提供理论依据。Objective: To explore the diagnostic role of miR-122 expression in glioma and its correlation with VEGF expression.Methods: Anticoagulant and serum samples were collected from 74 glioma patients and 74 healthy people.Quantitative reverse transcription PCR(qRT-PCR) was used to evaluate the diagnostic performance of miR-122 in glioma,and correalation analysis was conducted with the clinicopathological characteristics of glioma.The VEGF protein levels in serum of glioma patients and health people were quantitatively analyzed by enzyme-linked immunosorbent double antibody sandwich method(ELISA) and statistically analyzed.Results: The expression of miR-122 in glioma patients was significantly lower than that of health people.ROC analysis revealed high sensitivity and specificity of miR-122 for the diagnose of glioma(sensitivity 91.9% and specificity 81.8%,area under the curve0.939).The expression of miR-122 in glioma patients was significantly decreased with the progression of glioma,which was significantly correlated with WHO classification.The expression of VEGF was increased with the increase of glioma grade and the increase from Ⅲto Ⅳ was significant compared with the healthy control group.There was a negative correlation between miR-122 and VEGF.Conclusion: The expression of miR-122 was down-regulated significantly in glioma patients and it can be used as a biomarker for the diagnosis and prognosis.The decrease of miR-122 and the increase of VEGF provide theoretical basis for the occurrence and development of glioma.
