摘 要:目的:应用荧光成像技术考察磷酸铝凝胶(洁维乐~?)在小鼠胃肠道中的分布情况。方法:采用荧光探针Cy5标记洁维乐,经灌胃给药后在不同时间点对小鼠离体胃肠道进行荧光成像;同步监测小鼠的其他器官(心脾肝肾肺)的荧光分布。结果:口服Cy5标记物洁维乐后,在0.5 h时标记物已进入肠道内;2 h已分布至胃肠道各部位,且胃和小肠荧光信号开始减弱,4 h时盲肠和结肠的荧光信号增强,8 h后盲肠和结肠呈减弱趋势,然而在胃肠道停留时间可达24 h;同时,在小鼠其他器官内未显示荧光信号。结论:荧光探针Cy5可成功地对洁维乐进行胃肠道可视化示踪。Objective:To investigate the distribution of colloidal aluminumphosphate gel(Phosgel■)in gastrointestinal tracts of mice by fluorescence imaging.Methods:To visualize the distribution of Phosgel in gastrointestinal tracts,Cy5-marked Phosgel was administered to mice by oral gavage.The ex vivo fluorescence images of gastrointestinal tracts were taken at various time points after oral administration of Cy5-Phosgel.The ex vivo fluorescence images of other organs(heart,spleen,liver,kidney and lung)were also taken at the same time points.Results:At 0.5 h,Cy5-Phosgel appeared in the intestines;it was distributed in all parts of the gastrointestinal tracts at 2 h and then the fluorescence signals decreased in the stomach and small intestines.Subsequently,the fluorescence signals in cecum and colon started to increase at 4 h and to decrease at 8 h.In addition,there were still fluorescence signals in the gastrointestinal tracts at 24 h.However,no fluorescence signal was observed in other organs of the mice at the various time points.Conclusion:We successfully visualized the trace of Phosgel by fluorescence imaging and investigated its distribution and retention in gastrointestinal tracts of mice at different time points.
