摘  要:目的:评估多孔聚乳酸–羟基乙酸共聚物(PLGA)/Ⅱ型胶原(COLⅡ)软骨组织工程支架复合物的生物相容性。方法:通过低温快速成型技术(LDM)制备多孔PLGA支架,行低温等离子处理后复合COLⅡ,评估其力学性能。将多孔PLGA/COLⅡ软骨组织工程支架复合物与兔软骨细胞共培养,通过四甲基偶氮蓝比色法(MTT)测定检查细胞增殖,并通过扫描电子显微镜观察细胞与复合支架的黏附。结果:MTT检测实验组多孔复合支架细胞增殖与对照组比较,差异具有统计学意义(P <0.05);扫描电子显微镜显示兔软骨细胞在复合支架的表层和多孔结构中黏附、增殖和生长。结论:多孔PLGA/COLⅡ软骨组织工程复合支架可以适应软骨细胞的黏附和生长,具有良好的细胞相容性。Objective To evaluate the biocompatibility of porous PLGA/COL Ⅱ composite cartilage tissue engineering scaffold. Method Porous PLGA scaffolds were prepared by low–temperature deposition manufacturing(LDM), and type Ⅱ collagen was grafted on the surface of porous PLGA by low temperature plasma treatment. Evaluate the mechanical properties of composite scaffold. The porous PLGA/COL Ⅱ cartilage tissue engineering composite scaffold was cocultured with rabbit chondrocytes, and cell proliferation was examined by MTT assay, and the adhesion of the cells to the composite scaffold was observed by scanning electron microscope(SEM). Results MTT assay showed that the proliferation of porous composite scaffold co–cultured cells in the experimental group was significantly different from that in the control group(P < 0.05). SEM showed adhesion,proliferation and growth of rabbit chondrocytes on the surface and inside porous structure of the composite scaffold. Conclusion The porous PLGA/COL Ⅱ cartilage tissue engineering composite scaffold can adapt to the adhesion and growth of chondrocytes and has good biocompatibility.
