摘 要:目的:观察番木瓜叶提取物(Carica papaya leaf extracts,CPLE)对离体蛙心功能的影响,并初步探讨其作用机制。方法:采用斯氏蛙心插管方法,终质量浓度分别为0.4,0.8,1.6 g/L的番木瓜叶提取物对离体蛙心进行灌流,利用BL-420F生物信号采集系统记录其心率和心肌收缩力的变化。结果:CPLE对离体蛙心具有减缓心率和减小心肌收缩力的作用,随着CPLE浓度的增加,其作用逐渐明显,该实验最适药物质量浓度为0.8 g/L;去甲肾上腺素能缓解CPLE引起的心肌收缩力减弱作用,而CPLE亦能解除去甲肾上腺素诱导的负性肌力作用;乙酰胆碱和普萘洛尔可以在CPLE负性肌力作用的基础上进一步减小心肌的收缩力,反过来CPLE亦可在乙酰胆碱和普萘洛尔抑制心肌收缩力基础上加强其抑制作用。结论:CPLE可显著抑制离体蛙心心率和心肌收缩力,呈浓度依赖性。CPLE能够缓解NE诱导的负性肌力作用,且为乙酰胆碱、普萘洛尔的增效剂。Objective:to observe the effect of Carica papaya leaf extract(CPLE)on the isolated frog heart and explore its mechanism.Methods:the impact of different concentrations(0.4,0.8,1.6 g/L)of CPLE was investigated by frog heart perfusion,and the BL-420F biological signal acquisition system was used to record changes in heart rate and myocardial contractility.Results:CPLE was effective in slowing down heart rate and reducing myocardial contractility on isolated frog hearts.With the increase of the CPLE concentration,the effect became more apparent.The optimal concentration of CPLE was 0.8 g/L.Norepinephrine could alleviate CPLE-induced negative inotropic effect,and vice versa.Acetylcholine and propranolol could further reduce myocardial contractility on the basis of CPLE negative muscle strength,and vice versa.Conclusion:CPLE inhibited significantly both heart rate and myocardial contractility in a concentration-dependent manner.CPLE could mitigate NE-induced negative inotropic effect,and acetylcholine and propranolol could be used as synergists.
