摘 要:本研究旨在探究禽致病性大肠杆菌(APEC)中双组分系统phoP/Q与鞭毛Ⅲ型分泌系统(鞭毛T3SS)蛋白之间的联系。运用软件进行功能分析,预测phoP/Q与鞭毛T3SS的关系,利用基因芯片技术对APEC野生株和ΔphoP/Q缺失突变株进行转录组的差异性比较,筛选鞭毛T3SS基座中变化较大基因,并用荧光定量PCR检测基因表达变化量。结果显示:经STRING预测发现phoP/Q可能通过调控鞭毛T3SS基因座附近的双组分系统来间接调控鞭毛T3SS。基因芯片筛选与荧光定量PCR检测发现phoP/Q缺失能使鞭毛T3SS核心组件的大部分基因表达差异显著,同时也影响了双组分中motA、motB与cheY的表达。本研究对phoP/Q双组分系统与鞭毛T3SS的关系进行了预测与探索,为进一步研究揭示细菌的活动与致病机制提供了新的切入点与研究方向。The purpose of this study was to investigate the relationship between the two-component system phoP/Q and flagella typeⅢsecretory system(flagellar T3SS)proteins in Avian Pathogenic Escherichia coli(APEC).We used software to analyze the function and to predict the relationship between phoP/Q and flagella T3SS,and used gene chip technology to compare the transcriptome differences between APEC wild strain andΔphoP/Q deletion mutant,and screening for variable genes in flagellum T3SS pedestal.The changes of gene expression were detected by real-time PCR.The results predicted by STRING showed that phoP/Q may regulate flagellar T3SS indirectly by regulating the two-component system near the flagellar T3SS locus.Gene chip screening and real-time PCR revealed that the phoP/Q deletion could significantly differ the expression of most genes in the core component of flagellar T3SS,and also affect the expression of motA,motB and cheY in the two components.In this study,the relationship between phoP/Q two-component system and flagellar T3SS was predicted and explored,which provided a new breakthrough point and research direction for further research on bacterial activity and pathogenic mechanism.
