摘 要:为优化湖羊冻精生产工艺,提高冻精质量,以湖羊种公羊的精液为试验材料,分别选择7种稀释液进行处理,采用液氮熏蒸法进行冷冻,并设置不同解冻温度和时间进行交叉试验。结果表明:7号稀释液处理的湖羊精子在42℃和70℃两种解冻条件下的活力、活率、质膜完整率和顶体完整率均最高; 6种解冻条件中,70℃解冻5 s的解冻效果最佳,其活率、活力、质膜完整性和顶体完整率分别达50. 62%、43. 09%、40. 09%和54. 37%。综上,7号稀释液和70℃解冻5 s是湖羊精液冷冻的最佳稀释液和解冻条件。In order to optimize freezing process and improve the efficiency of Hu ram semen freezing,this study froze and thawed sperms using 0.25 mL straw,liquid nitrogen fumigation method under different defrost temperature and time with seven diluents(1-7)for cross test.The results showed that No.7 diluent got the highest motility,viability,plasma integrity and acrosome integrity in both 42 ℃ and 70 ℃ thawing conditions;70 ℃ 5 s got the best result in 6 thawing conditions.The motility,viability,plasma integrity and acrosome integrity of 70 ℃ 5 s group were 50.62%,43.09%,40.09% and 54.37%,which was higher than other groups.In conclusion,No.7 diluent was the optimal freezing diluent formulation and 70 ℃ 5 s was the best thawing condition.
