摘  要:选取平均体长为7. 8±1. 5 cm、平均体重为4. 45±0. 58 g的黄颡鱼幼鱼为试验对象。利用RT-PCR和RACE技术,测定NPY基因在黄颡鱼脑、肝、脾、肌肉4种组织中的相对表达量,并分析了在3种不同投喂策略下黄颡鱼幼鱼脑部NPY表达水平的变化,探讨百里香酚对黄颡鱼脑部NPY基因表达的影响。结果显示:黄颡鱼脑部的NPY基因表达量远远高于肝脏和脾脏,肌肉组织的NPY基因表达量最低。试验组Ⅰ在投喂基础饲料5 h后投喂百里香酚,NPY相对基因表达量出现明显上升(P <0. 05);试验组Ⅱ在投喂百里香酚5 h后,NPY相对基因表达量保持较高水平,没有明显变化,而在继续投喂基础饲料5 h后,其NPY基因表达量出现明显下降(P <0. 05)。以上结果表明百里香酚对黄颡鱼NPY基因表达有明显促进作用。An experiment was conducted to evaluate the effect of different strategy of the thyme of phenol on tissue s NPY expression of yellow catfish juvenile. Yellow catfish were chosen with average length 7.8±1.5 cm and average weight 4.45±0.58 g to evaluate the effect of expression in brain tissue under 3 different feeding strategies. The NPY mRNA expression in 4 tissues (brain, liver, spleen, muscle) was determined by RT-PCR and RACE. The results showed that the NPY gene expression in brain was significantly higher than other tissues, and NPY expression in muscle was lowest among all tissues. In group Ⅰ after rearing basal feed 5 h, the NPY expression of yellow catfish brain appeared significantly increased after rearing the thyme of phenol( P < 0.05);In group Ⅱ after rearing thyme 5 h, the NPY expression of yellow catfish brain was maintained at a higher level, but the NPY expression of yellow catfish brain had significantly declined after rearing basal feed 5 h( P <0.05). In conclusion the thyme of phenol has obviously promoted NPY gene expression of yellow catfish juvenile.
