摘 要:目的:研究优化构建高肺血流性肺动脉高压大鼠模型的制备方法,提高成模率。方法:将SD大鼠随机分为假手术组、传统分流组及优化分流组,每组25只。假手术组只进行开腹操作而不进行分流术,传统分流组采用传统分流术,优化分流组采用优化分流术。于术后11周,观察3组大鼠血管变化、右心室压力、肺动脉病理改变和右心室心肌肥厚指数,并比较传统分流组与优化分流组模型成模率。结果:优化分流组成模率为88%,高于传统分流组的48%,差异有统计学意义(χ~2=7.445,P=0.006)。苏木精—伊红(HE)染色显示:与假手术组相比,传统分流术组与优化分流术组大鼠中,可见肺动脉壁较假手术组明显增厚。优化分流组和传统分流组右心室收缩压、舒张压及右心室心肌肥厚指数均较假手术组明显增大(均P<0.05),而优化分流组与传统分流组间比较,差异无统计学意义(P>0.05)。结论:优化分流手术可成功制备高肺血流性肺动脉高压大鼠模型,并提高成模率,具有一定的推广应用价值。Objective: To optimize a rat model of pulmonary hypertension induced by high pulmonary blood flow. Methods: SD rats were randomly divided into a sham-operated group,a conventional group and an optimized group,with 25 rats in each group.The rats in sham-operated group were only treated to open the abdominal cavity,rats in conventional group received conventional shunt,and those in optimized group received optimized shunt.After 11 weeks of operation,vascular changes,right ventricular pressure,pathological changes of pulmonary artery and right ventricular hypertrophy index were observed.The success rate of conventional shunt and optimized shunt was compared. Results: The success rat of optimized shunt was 88%,which was higher than that of conventional shunt (48%)(χ^2=7.445, P=0.006).Hematoxylin-eosin (HE) staining showed that the pulmonary artery wall in conventional group and optimized group were significantly thicker than that in sham-operated group.The right ventricular pressure and right ventricular hypertrophy index were significantly increased in conventional group and optimized group compared with the sham-operated group (P<0.05).There was no significant difference between the conventional group and optimized group (P<0.05). Conclusion: Optimized shunt surgery could significantly improve the success rate of rat model of pulmonary hypertension induced by high pulmonary blood flow.
