摘 要:目的研究灵芝提取物(GLE)对小鼠酒精性肝损伤的保护作用及机制。方法将60只小鼠随机分为空白组、模型组、联苯双酯滴丸(BF)对照组及GLE低、中、高剂量组,每组10只。模型组灌胃50%乙醇(4 800 mg/kg),GLE高、中、低剂量组灌胃不同剂量的GLE(200、100、50 mg/kg),BF对照组灌胃联苯双酯滴丸溶液(300 mg/kg),空白组灌胃等体积生理盐水(0.2 ml/10 g)。每天灌胃1次,连续7 d。末次灌胃后2 h,空白组灌胃等体积生理盐水,其余组均灌胃50%乙醇(4 800 mg/kg)。禁食不禁水16 h后,眼球取血和摘除肝脏,检测血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)及肝脏中丙二醛(MDA)、超氧化物歧化酶(SOD)活性,并进行肝脏病理组织学检查。结果 GLE能降低急性肝损伤小鼠血清中ALT、AST含量,降低肝组织中MDA含量,提高肝组织中SOD活性,减轻肝损伤程度。结论 GLE对小鼠酒精性肝损伤有一定的保护作用,其作用机制可能与抗脂质过氧化作用有关。Objective To study the protective effect of Ganoderma Lucidum Extract(GLE) on alcoholic liver injury in mice and its mechanism. Methods 60 experimental mice were randomly divided into blank group,model group, bifendate(BF) control group and GLE high,medium and low dose groups,10 mice in every group.Model group was given 50% ethanol (4 800 mg/kg),GLE high, medium and low dose groups were given different doses of GLE (200,100,50 mg/kg),BF control group was given biphenyl diester drop solution (300 mg/kg),and blank group was given equal volume normal saline (0.2 ml/10 g).Intragastric administration was performed once a day for 7 consecutive days.2 hours after the last administration,normal saline was given to the blank group,and 50% ethanol was given to the rest groups(4 800 mg/kg). After fasting for 16 h,blood was taken from the eyeball and the liver was removed.The serum ALT,AST and SOD activities in the liver were detected,and the liver histopathological examination was performed. Results GLE can reduce the ALT,AST content in serum of mice with acute liver injury and the MDA content in liver tissue. It can also increase the activity of SOD in liver tissue and reduce the severity of liver damage. Conclusion GLE has protective effect on acute alcohol liver injury in mice and its mechanism may be related to its anti-lipid peroxidation.
