摘  要:目的探讨雌雄性豚鼠不同年龄自发性骨关节炎发生中关节软骨、软骨下骨显微结构的改变与体质量、性别的相关性。方法选用健康1月龄SPF级Hartley雌雄性豚鼠各18只,将不同性别的豚鼠随机分为3组,每组6只,分为1、3、5月龄组,取后肢膝关节。通过大体形态观察及X线检查评价关节软骨退变情况,采用Micro-CT观察胫骨软骨下骨显微结构的改变情况。结果雄性和雌性Hartley豚鼠的体质量随月龄增加而逐渐增加,3个月后雄性豚鼠体质量明显高于雌性豚鼠体质量。大体观察:5月龄雌雄性豚鼠胫骨平台、股骨髁、股骨髌面等关节面粗糙,光泽暗淡。X线检查:与1月龄相比,3月龄和5月龄雌雄性豚鼠膝关节间隙变窄。Micro-CT显示,3月龄和5月龄雌雄性豚鼠结构模型指数和各向异性的程度明显低于1月龄,而骨小梁厚度高于1月龄,内侧较外侧明显。结论随着年龄增加至5月龄的豚鼠自发性骨关节炎的发生可能与性别、体质量无关,对于软骨和软骨下骨两者的因果关系暂时难以确定。Objective To investigate the relationship of articular cartilage degeneration, change of subchondral bone ultrastructural, weight and sex by X-ray and Micro-CT in male and female guinea pigs with spontaneous osteoarthritis at different development stages. MethodsThirty-six (one month age;18 males and 18 females) Hartley guinea pigs were selected. They were divided into 3 groups according to different age and sex:one-month, 3-month and 5-month ( n =6 in a group). The knee joint of hind leg were collected. Articular cartilage degeneration in the knee joints was assessed by gross pathological examination and X-ray. The tibial subchondral bone was dissected and analyzed for ultrastructural changes by micro-computed tomography (micro-CT). ResultsThe weight of male and female Hartley guinea pigs increased with age and the male were significantly heavier than female guinea pigs after 3-month. The tibial plateau, femoral condyle and patellar fossa of femur were rough and dull at 5-month in the male and female guinea pig by the gross morphology . Compared with one-month, at the 3- and 5-month, the gap of knee joint was narrowed in male and female guinea pig by X-ray examination . At 3- and 5-month, the structure model index and degree of anisotropy were significantly lower than that one-month of male and female guinea pigs, but the trabecular thickness was higher than one-month. The medial was more obvious than lateral. ConclusionThe pathogenesis of spontaneous osteoarthritis with age increased to 5-month in male and female guinea pigs may have no relation with body weight and sexspecific difference, and the causality about cartilage and subchondral bone is difficult to confirm.
