摘 要:目的探讨槲皮素(Quercetin)对感染性休克大鼠肺脏的保护作用及其对TLR4/Myd88信号通路表达的影响。方法清洁级SD大鼠40只,体重250~300g,随机分为假手术组(Sham组)、感染性休克组(CLP组)、地塞米松阳性对照组(DEX组)和槲皮素处理组(Qu组),每组10只。采用大鼠盲肠结扎穿孔法建立大鼠感染性休克模型, Qu组于CLP后腹腔注射槲皮素;DEX组于CLP后腹腔注射地塞米松。发生后2h处死大鼠,观察大鼠肺脏组织病理学结果;血气分析结果;ELISA检测血清中TNF-α、IL-6、IL-10浓度;Western blot法检测TLR4、Myd88、NF-κB蛋白表达水平。结果槲皮素可以改善大鼠感染性休克的肺泡损伤, PaO_2和SaO_2值均升高,PaCO_2、 HCO_3值降低,血浆总TNF-α、IL-6降低,IL-10升高,且肺脏组织中TLR4、Myd88、NF-κB蛋白表达明显减低(与CLP组比较,P<0.05)。结论槲皮素可以有效抑制感染性休克导致的肺损伤,可能与调节TLR4/Myd88信号通路介导的炎症反应相关。Objective To study if TLR4/Myd88 signaling pathway is involved in the inhibition of quercetin on lung injury of rats with septic shock. Methods 40 SD rats were randomly divided into control group(Sham group), septic shock(CLP) and dexamethasone(DEX) and quercetin treatment group(Qu), 10 in each group. A rat model of septic shock was established by using cecal ligation and perforation. Qu group was injected with quercetin into the abdominal cavity after CLP. The DEX group was injected with dexamethasone after CLP. After the occurrence of 2h, rats were executed to observe the pathological results of lung tissue in rats. Serum TNF- alpha, IL-6 and IL-10 were detected by ELISA. Western-blot was used to detect the expression of TLR4, Myd88 and NF-.Κb. Results In CLP group, the alveolar damage was severe, and the pulmonary interstitial and alveolar cavity was severe edema, so a large number of inflammatory cell invasion and transparent film formation were observed;the values of PaO and SaO decreased, but elevated for PaCO and HCO;Serum 2 2 2 3 total TNF-alpha, IL-6 increased, IL-10 decreased, and the expression of TLR4, Myd88, and NF-κB in lung tissues increased compared with the Sham group. Quercetin inhibited the inflammatory response caused by septic shock, and the alveolar injury was significantly reduced, and the PaO and SaO values were all increased, and PaCO and HCO decreased. 2 2 2 3 Plasma total TNF- alpha, IL-6 decreased, IL-10 increased, and the expression levels of TLR4, Myd88, NF-κB protein in lung tissues decreased significantly (compared with CLP group, P< 0.05). Conclusion Quercetin can effectively inhibit lung injury caused by septic shock, which might be related to the inflammatory response mediated by TLR4/Myd88 signaling pathway.
