摘  要:目的观察雷帕霉素对大鼠脑缺血再灌注损伤的保护作用及对TLR4/MyD88/NF-κB信号通路的调节。方法 SD大鼠30只,随机分为三组,每组十只,分别为:假手术组(Sham组)、脑缺血再灌注损伤组(I/R组)和雷帕霉素预处理组(RAP组)。HE染色观察脑组织病理改变,TUNEL检测脑组织凋亡,ELISA检测脑损伤标志物和炎症因子的变化,Western blot检测脑组织TLR4、MyD88和NF-κB蛋白表达。结果雷帕霉素改善缺血再灌注造成的脑组织损伤,改善脑组织损伤造成的细胞凋亡,降低缺血再灌注损伤脑组织脑损伤标志物及炎症因子水平,雷帕霉素降低缺血再灌注损伤脑组织TLR4信号通路相关蛋白表达。结论雷帕霉素减轻大鼠脑缺血再灌注损伤与抑制TLR4/MyD88/NF-κB信号通路相关。Objective To observe the protective effect of Rapamycin on cerebral ischemia reperfusion injury in rats and its relationship with TLR4/MyD88/NF-κB signaling pathway. Methods 30 SD rats were randomly divided into sham group, cerebral ischemia reperfusion injury group(I/R group) and Rapamycin pretreatment group(RAP group). HE staining was used to observe histopathology changes in brain tissue. TUNEL assay was used to detect the apoptosis of brain tissue. ELISA was used to detect changes of brain damage markers and inflammatory factors. Western blot was used to detect the expression levels of TLR4, MyD88 and NF-κB proteins in brain tissue. Results Rapamycin improved ischemiareperfusion brain tissue damage, decreased tissue damage caused by cell apoptosis, reduced cerebral ischemia-reperfusion injury brain damage markers expression and inflammatory factor levels, and downregulted expression levels of TLR4 signaling pathways related proteins in brain tissue. Conclusion The alleviation of Rapamycin on the cerebral ischemia reperfusion injury was related to the downregulation of TLR4/MyD88/NF- B signaling pathway.
