摘 要:目的:研究柴金解郁颗粒剂的抗抑郁作用。方法:建立药物相互作用和应激行为作用诱导的两种小鼠抑郁模型和慢性应激大鼠抑郁模型,将动物随机分为空白组、模型组、阳性对照(盐酸氟西汀)组(0.06 g·kg^(-1))、柴金解郁颗粒剂低(0.13 g·kg^(-1))、中(0.26 g·kg^(-1))、高(0.52 g·kg^(-1))剂量组。检测小鼠体温、眼睑下垂次数、运动不能动物数、甩头次数、悬尾不动时间、强迫游泳不动时间;检测大鼠糖水偏嗜度、活动次数、学习记忆时间。结果:与模型组比较,柴金解郁颗粒各剂量对小鼠给药体温、眼睑下垂次数、运动不能、甩头次数、游泳不动时间(除柴金解郁颗粒低剂量组)和悬尾不动时间的影响差异有统计学意义(P<0.05或P<0.01),以上作用均呈剂量相关性,且部分剂量组与阳性对照组差异有统计学意义(P<0.05或P<0.01)。与模型组比较,柴金解郁颗粒各剂量对大鼠的糖水偏嗜度、总活动数、爬上平台时间和进入目标象限时间影响差异均有统计学意义(P<0.05或P<0.01),以上作用均呈剂量相关性,且高剂量组与阳性对照组效果相似。结论:柴金解郁颗粒剂对小鼠和大鼠的抑郁症状有明显的改善作用。Objective: To study the antidepressant effect of Chaijin Jieyu granules. Methods: Two kinds of mouse depression models induced by drug interaction and stress behavior were established,and a depression model of chronic stress was also established in rats. The animals were randomly divided into the blank group,the model group,the positive control group( fluoxetine hydrochloride,0. 06 g·kg^-1),and Chaijin Jieyu granules low( 0. 13 g·kg^-1),medium( 0. 26 g·kg^-1) and high( 0. 52 g·kg^-1) dose groups. The body temperature,eyelids drooping times,akinetic number,head shaking times,tail suspension time and immobility time in forced swimming were detected,and the sugar preference,activity frequency and learning and memory time were detected in rats.Results: Compared with the model group,different doses of Chaijin Jieyu granules showed significantly different effects on the body temperature,eyelid drooping times,akinetic number,head shaking times,swimming immobility time( except for Chaijin Jieyu granules low-dose group) and tail suspension time in mice( P < 0. 05 or P < 0. 01),the above effects were dose-related,and there was significant difference between the partial dose group and the positive control group( P < 0. 05 or P < 0. 01). Compared with the model group,different doses of Chaijin Jieyu granules showed statistically significant effects on the sugar preference,total activity,climbing platform time and entering target quadrant time in rats( P < 0. 05 or P < 0. 01),the above effects were dose-dependent,and the effects of high dose group were similar to those of the positive control group. Conclusion: Chaijin Jieyu granules can obviously improve the depressive symptoms of mice and rats.
