消瘤方对子宫内膜异位症患者子宫内膜细胞增殖和凋亡的影响

摘　　要：目的探讨中药消瘤方对子宫内膜异位症(EMS)患者在位和异位内膜基质细胞增殖和凋亡的影响。方法采用改良筛网法原代分离、培养EMS患者在位、异位及正常人子宫内膜基质细胞;将分离出的子宫内膜细胞分为3组,分别为正常组、在位组和异位组,其中在位组和异位组再随机分为生理盐水组、孕三烯酮组(西药组)以及消瘤方提取物5 g/L组(中药低剂量组)及消瘤方提取物10 g/L组(中药高剂量组),各组分别加入相应药物干预后,对内膜细胞的增殖以及凋亡状况进行分析。结果在使用药物干预后,西药组以及中药高剂量组可以起到抑制在位以及异位子宫内膜细胞增殖的作用,并且可以促进细胞的凋亡,并且差异具有统计学意义(P<0.05或0.01)。结论子宫内膜异位症的发生与EMS患者在位和异位内膜细胞的异常增殖和凋亡下降密切相关,消瘤方可通过抑制子宫内膜细胞的增殖和促进其凋亡的发生,从而阻止EMS的形成而起到治疗作用。Objective To investigate the effect of Chinese medicine Xiaoliu Decoction on proliferation and apoptosis of eutopic and ectopic endometrial stromal cells in patients with endometriosis(EMS). Methods The modified sieving method was used to separate and culture the eutopic, ectopic and normal human endometrial stromal cells of EMS patients. The isolated endometrial cells were divided into three groups, which were normal group, the eutopic group and the ectopic group,eutopic group and ectopic group were randomly divided into normal saline group, gestrinone group, Xiaoliu Decoction extract 5 g/L group and Xiaoliu Decoction extract 10 g/L group(high-dosage herb group), each group was added with corresponding drug intervention,and the proliferation and apoptosis of endometrial cells were analyzed. Results After drug intervention, western medicine group and high-dose Chinese medicine group could inhibit the proliferation of eutopic and ectopic endometrial cells, and promote cell apoptosis, and also the difference was statistically significant.(P<0.05 or 0.01). Conclusion The occurrence of endometriosis is closely related to the abnormal proliferation and apoptosis of eutopic and ectopic endometrial cells in EMS patients. Xiaoliu Decoction could inhibit the proliferation of endometrial cells and promote the occurrence of apoptosis, thereby preventing the formation of EMS and acting as a treatment.
