摘 要:从郑州市某公园土壤中筛选得到1株具有较好的异养硝化-好氧反硝化性能的细菌HJ-7。根据菌落及菌体形态、生理生化特征和16S rDNA序列的系统发育分析,对该菌株进行了鉴定,并研究了该菌的生长及异养硝化相关特性。初步鉴定该菌为施氏假单胞菌,命名为Pseudomonas stutzeri HJ-7,经过24 h的培养,该菌对氨氮和总氮的去除率可以分别达到90.8%和41%。利用菌株HJ-7建立同步硝化反硝化的生物脱氮系统,其脱氮性能进行研究,考察了温度、碳源、氨氮负荷和接种量等因素对脱氮效果的影响,并通过正交试验研究菌株最优脱氮条件。温度、碳源、氨氮负荷和接种量对HJ-7菌脱氮效果都有极其显著的影响(p<0.001);各因素对总氮去除效果影响程度依次为:温度>氨氮负荷>碳源>接种量;最适生长条件为:温度30℃、氨氮负荷40 mg/L、碳源为柠檬酸钠、接种量5%。An efficient heterotrophic nitrifying strain HJ-7 was isolated from a park in Zhengzhou, and identified to be the Pseudomonas stutzeri by its clone shape and 16S rDNA sequence. After 24 h culture, 90.8% ammonia nitrogen and 41% total nitrogen were removed by this strain. The effects of temperature, carbon source, ammonia nitrogen load and inoculums on the nitrogen removal characterization of this strain were investigated and an orthogonal experiment (4 factors and 3 levels) was designed on this basis, to study the the optimal nitrogen removal conditions. The orthogonal experiment results showed that the effect of temperature, carbon source , ammonia nitrogen load and inoculums on total nitrogen removal efficiency is extremely significant (p<0.001). The effect on the total nitrogen removal was in descending order as temperature>ammonia nitrogen load>carbon source>inoculums. The optimal nitrogen removal conditions were temperature 30℃ ammonia nitrogen load 40 mg/L, sodium citrate as carbon source and 5% inoculums.
