摘  要:目的:探究巨噬细胞Raw264.7氧化低密度脂蛋白(LDL)过程中肾素-血管紧张素系统(RAS)的变化。方法:培养RAW264.7细胞,LDL刺激后,用ACE活性测定试剂盒检测细胞内ACE活性的变化,用AngU试剂盒测定AngD的表达;Real-TimePCR检测血管紧张素转移酶(ACE)mRNA的表达;利用Real-TimePCR及WesternBlot检测血管紧张素U2型受体(AGTR2、AT2R)的表达。结果:LDL刺激Raw264.7细胞后,细胞内ACE的活性.AngH的表达、ACEmRNA水平、AT2RmRNA和蛋白质的表达都升高(P<0.05)。结论:L叽激活了肾素-血管紧张素系统。Objective To explore the changes of renin-angiotensin system (RAS) in the process of low-density lipoprotein (LDL) oxidation in Raw264.7 macrophage cells.Methods Raw264.7 cells were cultured and stimulated with LDL and then ACE activity was determined with Mouse ACE ELISA Kit, Anglfs expression was determined with Mouse Angll ELISA Kit;Angiotensin I-converting enzyme (ACE) was detected by Real-Time PCR, angiotensin II type 2 receptor (AGTR2, AT2R) was detected by Real-Time PCR and Western Blot Results After LDL stimulated Raw264.7 cells, the activity of ACE, AnglFs expression, ACE mRNA level, AT2R's expression were increased (P<0.05).Conclusion LDL activates renin-angiotensin system in Raw264.7 cells.
