摘  要:为探究鸡血管活性肠肽受体基因(VIP1和VIP2)和多巴胺受体基因(DRD1)多态性与性成熟的关系,采用PCR-SSCP和SNPs测序验证等方法对优质肉鸡F系287个个体进行了性成熟相关性状的关联分析。结果表明,VIP1基因处发现2个突变位点,分别为73352处C→T的突变,68818处T→C的突变;VIP2基因发现1个突变为36 127处A→G的突变。DRD1基因发现3个突变位点,分别为201处C→T的突变,208处A→G的突变及255处A→G的突变。与性成熟的关联性分析表明,VIP1基因73 352处的基因突变与冠高、冠长、冠重和睾丸重显著相关(P<0.05),而与生长激素GH、睾酮激素TEST差异不相关(P>0.05);VIP2和DRD1各处基因突变与性成熟相关指标均存在不同程度的差异显著(P<0.05),但都与生长激素GH和睾酮激素TEST差异不相关(P>0.05)。结果提示说明,VIP1、VIP2和DRD1基因可作为性成熟性状标记辅助选择的有效分子标记。This experiment was conducted to evaluate the relation of polymorphism of VIP1,VIP2 and DRD1 genes on sexual maturity in cocks,287 high quality broilers of F line were performed by PCR-SSCP and SNPs sequencing methods to be analyzed the association of polymorphisms sites with sexual maturity.The results showed that two mutation sites were detected in VIP1 gene,the bases C was changed to T on 73 352 site and bases C was changed to T on 68 818 sites respectively,one mutation site was detected in VIP2 gene,the bases A was changed to G on 36 127 sites.Three mutation sites were detected in DRD1 gene,the bases C was changed to T on 201 site and bases A was changed to G on 208 sites and bases A was changed to G on 255 sites respectively.Association analysis of sexual maturity production traits showed that comb height comb length and comb weigh spermary weight were correlated with gene mutation on73 352 sites of VIP1 gene(P<0.05),but was no correlated with GH content and TEST content(P>0.05).There were association correlated with VIP2 and DRD1 genes and sexual maturity in cocks of Broilers(P<0.05),and there were no association correlated with GH and TEST content(P>0.05).Based on these results,the VIP1,VIP2 and DRD1 genes may be regarded as an effective molecular marker on the marker assisted selection program for sexual maturity.
