摘  要:目的探讨脓毒症大鼠早期应用β受体阻滞剂对炎症介质的干预作用。方法选择健康雄性SD大鼠54只,随机分为假手术模型组、脓毒症组和β受体阻滞剂组,每组18只。各组造模3 h、6 h、18 h后麻醉处死大鼠,采用酶联免疫吸附法(enzyme linked immunosorbent assay, ELISA)检测血清TNF-α、IL-1β、IL-6,留取完整大鼠的左心、肺组织行病理切片,观察组织形态学变化。结果在术后各时间点β受体阻滞剂组血清TNF-α、IL-1β、IL-6水平均低于脓毒症组,差异有统计学意义(P <0.05);大鼠左心、肺组织病理组织形态学及透射电镜下细胞超微结构的损伤均小于脓毒症组。结论β受体阻滞剂应用于早期脓毒症大鼠,可干预炎症介质的释放及降低交感神经敏感性,对脏器具有保护作用。Objective To investigate the intervention effect of early application of P-blocker on inflammatory mediators in septic rats.Methods Fifty-four healthy male SD rats were randomly divided into three groups:sham group(n=18),sepsis group(n=18),β-blocker group(n=18).The rats were killed after 3 h,6 h and 18 h of anesthesia,and the level of serum TNF-α,IL-10,IL-6 were measured by enzyme linked immunosorbent assay(ELISA).The intact left ventricular myocardium and lung tissues of the rats were taken for pathological section and the histological changes were observed.Results The serum levels of TNF-α,IL-1β,IL-6 inβ-blocker group were lower than those in sepsis group,and the differences were statistically significant(P<0.05).The damages of left heart,lung histopathology and ultrastructure under transmission electron microscope inβ-blocker group were less than those in sepsis group.Conclusionsβ-blocker has protective effect on organ of early sepsis rats,which may be related to decreasing catecholamine sensitivity and inflammatory mediators released.
