摘 要:目的:探索黄连大黄肉桂复方及其有效组分配伍对人肝癌细胞株SMMC-7721端粒酶的影响。方法:将SMMC-7721细胞分为3组:对照组、复方组和组方组,分别给予0.1%二甲基亚砚(DMSO)、大黄黄连肉桂复方及其组分配伍(小檗碱、大黄素和桂皮醛)进行干预,作用24h后,在倒置相差显微镜下观察各组细胞数量及形态;收集细胞采用反转录-实时荧光定量聚合酶链反应(RT-qPCR)法检测端粒酶逆转录酶(hTERT)的mRNA表达;采用免疫印迹技术检测hTERT的蛋白表达。结果:黄连大黄肉桂复方及组方对人肝癌细胞株SMMC-7721的增殖具有一定的抑制作用,对hTERT的mRNA表达均有上调的趋势,而对hTERT的蛋白质表达具有下调的趋势,其中以组方组作用较甚。结论:黄连大黄肉桂复方及其有效组分可抑制人肝癌细胞株SMMC-7721的增殖,这可能与抑制hTERT的蛋白质表达有关。Objective:To explore the effect of Coptis chinensis-Rheum officinale-Cinnamon compound and its active ingredient combina-tion on human telomerase reverse transcriptase(hTERT)of SMMC-7721 human liver cancer cell line. Methods:SMMC-7721 cells were divid- ed into three groups that were control group,the compound group and the composition group,and which were treated with 0. 1% DMSO,Coptis chinensis-Rheum officinale-Cinnamon compound and its active ingredient combination (BBR,emodin and cinnamaldehyde) respectively. Af- ter treatment for 24h,the growth and morphology of the cells in each group were observed under the inverted phase contrast microscope,the mRNA expression of hTERT was detected by RT-qPCR and the expression of protein of hTERT was measured by Western blot. Results:The Coptis chinensis-Rheum officinale-Cinnamon compound and its active ingredient combination inhibited the proliferation of SMMC-7721 cells, up-regulated the mRNA expression of hTERT and down-regulated the expression of protein of hTERT slightly. Thereinto,the composition group was more effective. Conclusion:The effect of Coptis chinensis-Rheum officinale-Cinnamon compound and its active ingredient combina- tion on inhibting the proliferation of SMMC-7721 cells maybe related to the suppression of the protein expression of htert.
