摘  要:为了深入研究玉米弯孢霉叶斑病,从三亚南滨农场等南繁种植基地采集病叶,分离得到致病力强的玉米弯孢霉叶斑病病菌C24。利用柯赫氏法则和分子生物学鉴定、生长速率测定等方法 ,分别从培养基(PSA、PDA、 Czapek、 PSA+玉米绿叶煎汁、玉米绿叶煎汁、 PDA+玉米绿叶煎汁、燕麦)、温度(10、 15、 20、 25、 28、33、 37、 41℃)、光照(全日光照、全日黑暗)、 pH(5、 6、 7、 8、 9、 10、 11)、碳源(蔗糖、麦芽糖、乳糖、 D-木糖、 D-果糖、可溶性淀粉)、氮源(甘氨酸、苏氨酸、 NaNO3、 L-缬氨酸、 L-丝氨酸、 L-精氨酸、 L-谷氨酰胺、 L-赖氨酸、 L-脯氨酸、 L-亮氨酸)6个方面对南繁区玉米弯孢霉叶斑病菌C24的生物学特性进行了研究。结果显示:C24鉴定为弯孢菌(Curvularia lunata Boedijn)在供试培养基上均能生长,其中在燕麦培养基上生长最好;最适生长温度在28~37℃;最适pH值为7~9;光照有利于菌丝生长;最适碳源为麦芽糖,氮源为L-亮氨酸。本研究发现玉米弯孢霉菌能引起南繁区玉米病害,较适应热带地区高温天气,在28~37℃生长旺盛。Leaves of corn infected with leaf spot disease were collected from the corn field in Nanbing State Farm,Sanya for further study of the leaf spot disease caused by Curvularia lunata Boedijn,and a strain C24 of the leaf spot disease was isolated.This isolate was determined by using Koch's rule,mycelium growth method and molecular bioassay.The isolate was cultured on different mediums (PDA medium,PSA medium,corn leaves juice medium,PDA+corn leaves juice,PSA + corn leaves juice,oatmeal medium,Czapek medium) under different conditions of temperature (10,15,20,25,28,33,37,41℃),illumination (light,dark),pH (5,6,7,8,9,10,11),carbon source (sucrose,maltose,lactose,D- xylose,D- fructose,soluble starch) and nitrogen sources (L-arginine,L-serine,L-lysine and L-glutamine,L- leucine,L-proline,L-valine,threonine,glycine,NaNO3).Results show that the ribosomal DNA-ITS sequence of C.lunata Boedijn had a 99% similarity with that in the GenBank database.Therefore,the strain C24 isolated from the corn leaf spot disease in the south of Hainan was C.lunata Boedijn.The strain C24 could grow on all the culture mediums and grew best under the cultural conditions of oatmeal medium as cultural medium,temperature at between 28℃ and 37℃,pH 7-9,light culture,maltose as carbon source,L leucine as nitrogen source.Culture under the light facilitated mycelium growth.This study found that C.lunata Boedijn could cause the corn leaf spot disease in the south of Hainan and was suitable for growth at high temperature in the tropics,especially at between 28℃ and 37℃.
