摘 要:目的探讨太白山藤梨根提取物对人喉癌细胞HeP-2的生长抑制作用。方法采用细胞计数法统计人喉癌细胞HeP-2的增殖率,采用MTT(四甲基噻唑蓝)法测定太白山藤梨根提取物对HeP-2细胞的剂量-效应曲线、时间-效应曲线。采用Real-time PCR检测太白山藤梨根提取物作用HeP-2细胞后,COX-2、P65、VEGF mRNA的表达。结果太白山藤梨根提取物作用于HeP-2细胞后,随着药物浓度的增加,HeP-2细胞存活的百分率呈下降趋势,表现为明显的浓度依赖性。太白山藤梨根提取物各浓度作用时间在12、24、48、72、96、120 h时,表现出明显的剂量依赖关系和时间依赖关系,随着作用时间的延长和药物浓度的增加,HeP-2细胞的存活率呈明显下降趋势。太白山藤梨根提取物作用HeP-2细胞后,随着作用时间的延长,COX-2 mRNA、P65 mRNA、VEGF m RNA的表达均受到不同程度的抑制。各作用时间的COX-2 mRNA、P65 mRNA、VEGF mRNA表达均差异有统计学意义(P<0.01)。结论太白山藤梨根提取物能够抑制人喉癌细胞HeP-2的增殖,这种抑制作用具有时间依赖性和剂量依赖性,其抑制作用可能与下调COX-2、P65、VEGF的表达密切相关。Objective To investigate the inhibitory effect of Taibaishan vine root extract on the growth of human laryngeal carcinoma cell line HeP-2. Methods The proliferation rate of human laryngeal carcinoma cell line HeP-2 was counted by cell counting method. The dose-effect curve and time-effect curve of Hep-2 cells in Taibaishan vine root extract were de-termined by MTT (tetramethylthiazolyl blue) method. The expression of COX-2, P65 and VEGF mRNA was detected by Re-al-time PCR after detection of Hep-2 cells in Taibaishan Fujisui root extract. Results After the extract of Taibaishan vine root extract was applied to HeP-2 cells, the percentage of HeP-2 cells surviving decreased with the increase of drug con-centration, showing a significant concentration-dependent. At the 12, 24, 48, 72, 96 h, and 120 h, the concentration of the extract of Taibaishan vine root extract showed obvious dose-dependency and time-dependent relationship. With the prolon-gation of the action time and the increase of drug concentration, and the survival rate of HeP-2 cells showed a significant downward trend. After treatment with HeP-2 cells, the expression of COX-2 mRNA, P65 mRNA and VEGF mRNA were in-hibited in different degrees. The expressions of COX-2 mRNA, P65 mRNA and VEGF mRNA were significantly different (P<0.01). Conclusion The extract of Taibaishan vine root can inhibit the proliferation of human laryngeal carcinoma cell line HeP-2. The inhibition is dose-dependent and time-dependent, which may be closely related to the down-regulation of COX-2, P65 and VEGF expression.
