摘 要:目的探讨新型川芎嗪-香豆酸衍生物(T-CA)对拟脑缺血模型的形态保护作用。方法通过体外培养,使用神经生长因子(NGF)诱导分化PC12细胞,预先给予一定浓度(60μmol·L^(-1))的新型川芎嗪-香豆酸衍生物保护后,加入氯化钴(CoCl_2,300μmol·L^(-1))损伤细胞。采用Giemsa染色、DAPI染色、吖啶橙(AO)及吖啶橙/溴化乙锭(AO/EB)染色等方法,观察T-CA对CoCl_2损伤PC12细胞模型的形态保护作用。同时观察T-CA对大脑中动脉缺血(MCAO)模型大鼠的存活情况及脑皮层神经细胞形态的影响。结果与损伤组相比,给药组可显著抑制CoCl_2损伤PC12细胞的凋亡,缓解其形态损伤并提高MCAO模型大鼠的存活率。结论新型川芎嗪-香豆酸衍生物T-CA对CoCl_2损伤PC12细胞模型具有显著的神经保护作用,可提高MCAO模型大鼠的存活率。Objective To study the neuroprotective effect of novel ligustrazine- coumaric acid derivative (T-CA) on the morphology of cerebral ischemia model. Methods PC12 cells were cultured and differentiated by nerve growth factor (NGF) and were pre-protected by T-CA (60 μmol ·L -1 ) before the treatment with CoCl 2 (300 μmol·L^-1 ) as an trauma model in vitro .Giemsa staining,DAPI staining,acridine orange(AO) staining and AO/EB staining were used to study the neuroprotective effect of T-CA on morphologic appearance of CoCl2-induced neurotoxic PC12 cell model.Meantime,effects of T-CA on the survival condition of middle cerebral artery occlusion(MCAO) model and morphological change of rats' cortical neuron were observed either.Results The treatment group could significantly alleviate the morphological damage of CoCl 2 -induced neurotoxic PC12 cells model and improve the survival rate of MCAO model rats. Conclusion The results showed that the novel ligustrazine derivative T-CA had a significant neuroprotective effect on CoCl 2 -induced neurotoxic PC12 cells model and could enhance the survival rate of MCAO rats at the same time.
