摘  要:目的观察在人肝癌细胞系脂肪代谢过程中,腺瘤样结肠息肉基因(APC)对细胞线粒体的影响。方法 APC基因敲减质粒转染人肝癌细胞(BEL-7402)。用油酸刺激各组细胞,使细胞产生脂质堆积。油红O染色法检测各组细胞脂质堆积情况;使用ATP含量检测试剂盒检测各组细胞ATP含量;使用荧光探针标记法检测各组细胞内氧自由基(ROS)的含量。结果给细胞转染质粒后,APC基因的mRNA表达量降低(P<0.05),蛋白表达也降低(P<0.05);与对照组细胞相比,APC基因敲减组细胞加油酸刺激后细胞脂质堆积减少(P<0.05),ATP含量增高(P<0.05),细胞内氧自由基数量降低(P<0.01)。结论 APC在人肝癌细胞系BEL-7402脂质代谢过程中影响线粒体的生理功能。Objective To observe the effect of adenomatous polyposis coli (APC) on cell mitochondria in the process of fat metabolism in human hepatoma cell line. Methods Human hepatoma cell line(BEL-7402) cells were transfected with APC gene knockdown plasmid, and oleic acid was used to stimulate knockdown cells and control cells to produce lipid accumulation. Oil Red O staining was used to detect the lipid accumulation in each group. ATP content detection kit was used to detect the ATP content of each group. The content of oxygen free radicals (ROS) in each group was detected by fluorescent probe labeling. Results After transfection of the plasmid, both the mRNA expression( P <0.05) and protein expression of APC decreased ( P <0.05). Cell lipid accumulation after stimulation with oleic acid in APC knockdown group reduced as compared with control cells ( P <0.05), ATP content increased ( P <0.05), and the quantity of intracellular oxygen free radicals decreased ( P <0.01). Conclusions APC affects the physiological function of mitochondria during lipid metabolism in human hepatoma cell line BEL-7402.
