摘 要:吡咯是许多具有生物活性的天然产物和药物活性成分的基本结构单元,合成吡咯最经典的方法是Paal-Knorr反应。在微通道反应器中考察Paal-Knorr反应的动力学,回归反应速率常数。实验以2,5-己二酮与乙醇胺为原料,甲醇为溶剂,盐酸甲醇溶液为淬灭剂,合成1-(2-羟基乙基)-2,5-二甲基吡咯,通过气相色谱仪(GC)对反应进行定性、定量分析。随后对反应条件(反应温度、保留时间)进行优化,得到最佳反应条件:反应温度150℃,保留时间37.1 s。在确定最佳反应条件的前提下,进行反应动力学的拟合,最终确定在75℃下,该反应为二级反应,与釜式反应级数相一致,其反应速率常数为0.0308 L^(-1)·mol^(-1)·s^(-1)。Pyrrole are the basic building blocks of many biologically active natural products and active pharmaceutical ingredients,the most classic method for synthesis of pyrrole is the Paal-Knorr reaction. The kinetics of the Paal-Knorr reaction was investigated in a microchannel reactor,and the reaction rate constant was regressed. The synthesis for 1-(2-hydroxyethyl)-2,5-dimethylpyrrole is analyzed qualitatively and quantitatively by gas chromatography(GC),while the 2,5-hexanedione and ethanolamine are used as raw materials,methanol is used as solvent and methanol hydrochloride solution is used as quencher. Subsquently,the optimum reaction condition ,reaction temperature 150 ℃,retention time 37.1 s,was obtained after optimizations on the reaction conditions (including reaction temperature and retention time). Under optimum reaction condition patameters,further exploration of reaction kinetics was carried out,Finally,it is confirmed that the reaction is a second order reaction under 75℃,which is consistent with the batch reaction order,and the reaction rate constant is 0.0308 L^-1·mol^-1·s^-1.
