摘  要:[目的]确定黄芪药效成分中3种主要活性成分闪式提取的最佳工艺条件。[方法]分别采用硝酸铝-亚硝酸钠法、苯酚-硫酸法、香草酫-硫酸法对黄芪黄酮、多糖、皂苷的含量进行测定,以各成分得率为指标,采用响应面法对提取工艺参数进行优化并得到回归模型。[结果]结果表明:回归模型较好地反映了黄酮、多糖、皂苷得率与提取条件的关系,最优工艺条件分别为,黄酮∶乙醇浓度85%、料液比1∶25、提取时间60s、提取电压120V;多糖:乙醇浓度2.5%、料液比1∶15、提取时间105s、提取电压115V;皂苷:乙醇浓度70%、料液比1∶30、提取时间120s、提取电压110V。[结论]此工艺条件下提取黄酮、多糖、皂苷的得率为0.663%、0.965%、1.126%,回归模型的预测值与实测值的相差均小于0.05%,3个回归方程与实际情况拟合较好,优化工艺可靠。[Objectives] The aim of current study was to determine the optimal technological conditions for the flash extraction of three main active components in Astragalus membranaceus .[Methods] The contents of flavonoids, polysaccharides, and saponins were determined by ultraviolet, aluminum nitrate-sodium nitrite method, phenol-sulfuric acid method, and vanilla-sulfuric acid method, respectively. The yield of each component was used as the indexes to optimize the extraction process parameters by response surface methodology, and the regression models were obtained.[Results] The results showed that the developed regression model precisely reflected the relationship between the yield of flavonoids, polysaccharides, and saponins and extraction conditions. The optimal technological conditions were determined as following: the higher concentration of flavonoids was obtained when ethanol concentration at 85 %, solid-liquid ratio at 1:25, extraction time at 60 s, and extraction voltage at 120 V;the highest concentration of polysaccharides was achieved when ethanol concentration at 2.5 %, solid-liquid ratio at 1 ∶15, extraction time at 105 s, and extraction voltage at 115 V;while the greatest concentration of saponin was reached when ethanol concentration at 70 %, solid-liquid ratio at 1 ∶30, extraction time at 120 s, and extraction voltage at 110 V.[Conclusion] Under the mentioned conditions, the yield of flavonoids, polysaccharides, and saponins were 0.663%, 0.965%, and 1.126%, respectively. The difference between the predicted and measured values of the regression models were less than 0.05%. The three regression equations fitted well to the actual situation and the optimized process was reliable.
