摘  要:目的探讨盐酸法舒地尔对治疗脑动脉瘤模型大鼠的治疗效果及对血清肿瘤坏死因子α(TNF-α)、转化生长因子β1(TGF-β1)和血管内皮生长因子(VEGF)表达的影响。方法选择SD健康雌性大鼠30只,随机分为正常组、模型组和实验组,每组10只。对模型组、实验组大鼠进行脑动脉瘤模型建立,正常组不做处理。实验组大鼠注射盐酸法舒地尔,模型组和正常组大鼠注射等量0.9%氯化钠溶液,7 d后对大鼠脑动脉瘤血管壁厚度、肿瘤体积进行比较,HE染色后在显微镜下观察大鼠病理学组织,酶联免疫吸附试验法检测血清TNF-α、TGF-β1、VEGF水平,Western blot检测Rho激酶、NF-κBp65表达水平。结果模型组大鼠血管壁厚度、肿瘤体积高于正常组,实验组大鼠血管壁厚度、肿瘤体积显著低于模型组(P<0.05)。模型组大鼠血清TNF-α、TGF-β1、VEGF表达水平高于正常组,实验组大鼠血清TNF-α、TGF-β1、VEGF表达水平显著低于模型组(P<0.05)。模型组Rho激酶、NF-κBp65水平显著高于正常组,且实验组Rho激酶、NF-κBp65水平低于模型组,差异有统计学意义(P<0.05)。结论盐酸法舒地尔对脑动脉瘤的治疗效果显著,能够有效降低血清TNF-α、TGF-β1、VEGF的表达水平,为临床脑动脉瘤的治疗和预后提供重要的理论依据。Objective To investigate the therapeutic effect of fasudil hydrochloride on cerebral aneurysm in model rats and its effects on serum tumor necrosis factor α(TNF-α),transforming growth factor beta 1(TGF-β1) and vascular endothelial growth factor(VEGF) expression.Methods Thirty healthy female SD rats were randomly divided into normal group,model group and experimental group,with 10 rats in each group.The model of cerebral aneurysm was established in model group and experimental group.The rats in experimental group were given fasudil hydrochloride by injection,and the rats in model group and normal group were injected with equal amount of 0.9% sodium chloride solution.After 7 days,the vascular wall thickness of cerebral aneurysm and tumor size were observed and compared.After HE staining,the changes of pathological organization were observed under microscope,moreover the enzyme-linked immunosorbent assay method was used to detect the serum levels of TNF-α,TGF-β1,VEGF,and Western Blot was used to detect the expression levels of Rho kinase,NF-κBp65.Results The vascular wall thickness of cerebral aneurysm and tumor size were significantly higher than those in normal group,while the thickness of blood vessel wall and tumor volume in experimental group were significantly lower than those in model group(P<0.05).The expression levels of TNF-α,TGF-β1 and VEGF were in model group were significantly higher than those in normal group,which in experimental group were significantly lower than those in model group(P<0.05).The expression levels of Rho kinase and NF-κBp65 in model group were significantly higher than those in normal group,which in experimental group were significantly lower than those in model group(P<0.05).Conclusion The fasudil hydrochloride can inhibit the invasion and metastasis of cerebral aneurysm,which can effectively reduce the expression levels of serum TNF-α,TGF-β1 and VEGF,which provides important theoretical basis for the treatment and prognosis of clinical middle cerebral aneurysms.
