甘草-马齿苋配伍对特应性皮炎的免疫调节作用及其机制研究

摘　　要：目的:本院应用甘草和马齿苋湿敷治疗湿疹在临床取得了较好的疗效,但其具体作用的机制尚不明确。在中药的配伍研究中,药物配伍后的终浓度也和实验结果有直接关系。研究设计了两组不同浓度的配伍组,即配伍低浓度(减半配伍)和配伍高浓度(不减半配伍),考察药物配伍前后浓度对药效的影响。方法:通过特应性皮炎(AD)小鼠模型,初步探讨甘草和马齿苋对AD的免疫调节作用及可能的分子机制。采用药效学分析以及HE染色、酶联免疫吸附法ELISA、半定量RT-PCR等方法分别检测甘草和马齿苋对AD相关细胞因子、转录因子表达的影响。结果:局部应用甘草和马齿苋能够抑制AD小鼠皮肤增厚、脾脏指数、炎症细胞浸润以及降低血清IgE、IL-4、IL-6的含量和降低皮损中IL-4、IL-6、IL-13、IFN-γ炎症因子的mRNA的表达。结论:甘草和马齿苋能够通过抑制炎症反应和调节AD小鼠免疫功能,调节机体的Th1/Th2失衡发挥作用,两者配伍应用效果优于单独应用,并与配伍浓度有关,既配伍高浓度组优于配伍低浓度组。Objective The application of Glycyrrhizae Radix(GR) and Portulacae Herb(PH) in the treatment of eczema has achieved good clinical results, but the mechanism of its specific effects is still unclear. In addition, in the compatibility study of traditional Chinese medicine, the final concentration after drug compatibility is also directly related to the experimental results. Therefore, this study designed two different concentrations of compatibility groups, low concentration and high concentration. Methods AD was induced by 2, 4-Dinitrochlorobenzene(DNCB). The effects of GR and PH on the expression of AD-related cytokines and transcription factors were detected by pharmacodynamic analysis, HE staining, ELISA assay and semi-quantitative RT-PCR. Results GR and PH decreased skin thickness and spleen index of mice. They reduced the levels of IgE, IL-4 and IL-6 in mice serum. They down-regulated mRNA expression of IL-4, IL-6, IL-13 and IFN-γin mouse skin tissue. Conclusion GR and PH could exhibit suppressive effects on inflammatory response and immune function of AD, which indicates the possibility of developing AD as a useful drug for the treatment of AD.
