摘  要:对89株临床分离株采用PCR产物克隆测序和成簇规律间隔短回文重复序列(CRISPR)系统生物信息学分析等方法研究鲍曼不动杆菌CRISPR系统对靶基因的调控作用。结果表明:2株分离株(AB43与ATCC19606)具有CRISPR系统,它们间区靶向的基因有IV型分泌蛋白Virb5、带状黏附毒素蛋白、噬菌体蛋白、DNA聚合酶等。CRISPR系统抑制Virb5、带状黏附毒素蛋白基因的编码与表达,可能对鲍曼不动杆菌的生物膜形成具有一定调控作用。PCR cloning, sequencing and CRISPR analysis were performed in 89 clinical isolates to explore the regulation of A?baumannii CRISPR system on its target genes. The results showed that the two strains, AB43 and ATCC19606, contained a CRISPR system respectively. The genes targeted by them included type IV secreted protein Virb5, zonular occludens toxin protein, phage protein, DNA polymerase, etc. The inhibition of CRISPR system on the expression of Virb5 and zonular occludens toxin genes indicated a certain regulation of the biofilm formation of Acinetobacter baumannii.
