摘 要:目的制备一种以凝溶胶蛋白(gelsolin,GSN)为靶点载紫杉醇(paclitaxel,PTX)的PLGA (GSN-PTX-PLGA)微球,并研究其对Hca-F荷瘤小鼠体内治疗效果及抑制淋巴结转移的作用。方法采用4~6周龄雌性615小鼠建模将Hca-F细胞植入小鼠足垫,1周后观察小鼠成瘤情况。设对照组(生理盐水组)、PLGA微球组、PTX注射液组、PTX-PLGA微球组和GSN-PTX-PLGA微球组。分别对Hca-F细胞荷瘤小鼠尾静脉注射药物进行治疗、观察记录并绘制肿瘤体积生长曲线,给药4周后观察各组小鼠淋巴结转移情况。结果给药4周后GSN-PTX-PLGA微球组与PTX-PLGA微球组肿瘤生长曲线平缓,肿瘤体积较小与对照组比较(P<0.05),GSN-PTX-PLGA微球组小鼠淋巴结转移率(30%)与其他各组相比显著降低。结论 GSN-PTX-PLGA微球组与PTX-PLGA微球组对Hca-F细胞荷瘤小鼠的治疗有明显抑瘤效果,GSN-PTX-PLGA微球组对淋巴结转移有一定的抑制作用。Objective To prepare a kind of gelsolin-targeted PLGA microspheres with paclitaxel loaded and to evaluate the treatment effect and the lymph node metastasis of Hca-F tumor-bearing mice.Methods The model of 615 female mice aged 4-6 weeks were used to implant the Hca-F cells into the mouse foot pad.The tumor growth of the mouse footpads were observed one week later.And the tail vein injection was done by using physiological saline,PLGA microspheres,paclitaxel,paclitaxel loaded PLGA(PTX-PLGA)microspheres and Gelsolin-targeted,paclitaxel loaded PLGA microspheres(GSN-PTX-PLGA).After intravenous injection of drugs for tumor formation in mice with Hca-F cells,the tumor growth curve was recorded and the lymph node metastasis of each group was observed after four weeks of treatment.Results After 4 weeks of treatment,the tumor size and volume in the group treated with GSNPTX-PLGA microspheres and PTX-PLGA microspheres were obviously decrease compared with other groups(P<0.05),and the lymph node metastasis rate in the group treated with GSN-PTX-PLGA microspheres was significantly less than those of other groups.Conclusions The GSN-PTX-PLGA microspheres and the PTX-PLGA microspheres have a significantly inhibitory effect on the tumors in the Hca-F cell bearing mice,and the GSN-PTX-PLGA microspheres has a certainly inhibition effect on lymph node metastasis.
