摘 要:目的探讨大鼠硅胶植入后纤维包膜中巨噬细胞数量及表型变化。方法将30只大鼠分为第1天组、第3天组、第7天组、第14天组、第35天组5个组,每组6只。于大鼠背部肉膜下植入10 ml硅胶扩张器,术后上述时间点取材切片行免疫荧光双染色,检测各个时间点包膜中巨噬细胞数量以及M1/M2型巨噬细胞随时间占比变化。结果CD68^+细胞在第1天每个视野下细胞数(65.8±12.9)个明显小于第3天(102.8±14.5)个、第7天(116.8±14.2)个,差异有统计学意义(P<0.05);第7天每个视野下CD68数量明显多于第14天(56.8±12.9)个以及第35天(21.4±6.35)个,差异有统计学意义(P<0.05);iNOS^+CD68^+M1型巨噬细胞占比第1天0.48±0.13,第3天0.60±0.13,均大于第7天0.21±0.03,第14天0.17±0.03,第35天0.17±0.04,差异有统计学意义(P<0.05);CD206^+CD68^+M2型巨噬细胞第1、3、7、14、35天占比分别为0.70±0.06、0.60±0.07、0.70±0.08、0.67±0.02、0.60±0.06,各组间比较,差异无统计学意义(P>0.05)。结论大鼠巨噬细胞在硅胶植入后一直存在,早期M1、M2型巨噬细胞比例增加,后期以M2型巨噬细胞为主。Objective To study the temporal distribution of macrophage and its phenotype markers in fibrous capsules around silicone implants.Methods Thirty rats were randomly divided into five groups:days 1,3,7,14 and 35.Silicone prostheses(10 ml)were implanted subcutaneously into backs of rats.On each indicated day,the tissue specimens were collected,fixed in 4%paraformaldehyde for 24 hours and embedded in paraffin.Immunofluorescence was used to detect temporal distribution of M1/M2 macrophages.Results The number of CD68+macrophages at day 1(65.8±12.9)was smaller than that at day 3(102.8±14.5,P<0.05)and day 7(116.8±14.2,P<0.05);and the number of CD68+macrophages at day 7 was larger than that at day 14(56.8±12.9,P<0.05)and day 35(21.40±6.35,P<0.05);the proportion of iNOS^+CD68^+M1 cells at day 1 and day 3 was 0.48±0.13,0.60±0.13,respectively,and they were higher than that at day 7(0.21±0.03,P<0.05),day 14(0.21±0.03,P<0.05)and day 35(0.17±0.04,P<0.05);the proportions of CD206^+CD68^+M2 cells at day 1,day 3,day 7,day 14,day 35 were 0.70±0.06,0.60±0.07,0.70±0.08,0.67±0.02 and 0.60±0.06,respectively.Conclusions After the implantation of silicone prostheses,M1 cells increase in early stages and M2 cells maintain in high level throughout the experiment period.
