摘  要:目的:探讨不同剂量黄连降糖片对链脲佐菌素(STE)诱导的2型糖尿病模型大鼠氧化应激反应的影响及其量效关系。方法:将SPF级SD雄性大鼠120只随机分为正常组(n=10)和造模组(n=110),将造模组高脂高糖饲料喂养加小剂量链脲佐菌素腹腔注射建立SD大鼠2型糖尿病模型后随机分为模型组、黄连降糖片组(1~9片)及阳性组,每组10只。运用蒸馏水、黄连降糖片及盐酸二甲双胍分别干预4周后检测各组大鼠血清谷胱甘肽过氧化物酶(GSH-Px)活性、超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量。结果:与模型组相比,黄连降糖片2~9片组大鼠空腹血糖(FBG)降低(P<0.05),血清GSH-Px、SOD活性增强(P<0.05),MDA含量降低(P<0.05),与阳性组相比,黄连降糖片3~9片组大鼠血清GSH-Px活性增强(P<0.05),5~9片组大鼠血清SOD活性升高(P<0.05),6~9片组大鼠血清MDA含量降低(P<0.05),且在2~8片范围内随着药物剂量的增加,降糖幅度增加,调节氧化应激的作用增强。结论:黄连降糖片的降糖机制可能与抑制2型糖尿病模型大鼠氧化应激反应有关,且在一定剂量范围内存在量效关系。Objective: To explore different doses of HuangLian JiangTang tablets on oxidative stress reaction of rat model with streptozotocin (STE)-induced type two diabetes mellitus (T2DM) and the dose-effect relationship. Methods: A total of 120 male rats were randomized into the normal group (n=10) and the modeling group (n=110), the modeling group accepted high fat and high glucose feed and intraperitoneal injection of small dose of STE to establish rat models with T2DM, which were randomized into the model group, HuangLian JiangTang tablets groups (one to nine tablets), positive group, ten rats each group. The contents of MDA, the activity of SOD and GSH-Px in the serumof the rats in different groups were detected after treating for four weeks. Results: Compared with the model group, FBG of the rats in HuangLian JiangTang tablets group of two to nine tablets decreased (P<0.05), the activity of GSH-Px and SOD enhanced(P<0.05), the contents of MDA lowered(P<0.05), compared with positive group, the activity of GSH-Px in HuangLian JiangTang tablets group of three to nine tablets was notably higher than that of the positive group (P<0.05), the activity of SOD in the group of five to nine tablets were obviously higher than that of the positive group (P<0.05), the contents of MDA in the group of six to nine tablets were notably lower than these of the positive group (P<0.05), the scope of lowering glucose raised and the effects of regulating oxidative stress enhanced as the dose increased in the range between two and eight tablets. Conclusion: The glucose-lowering mechanism might be related to inhibiting oxidative stress reaction of T2DMrat models, and the dose-effect relationship exists in a certain dose range.
