摘 要:试验选取菌株PY-M3和PY-F6所产絮凝剂,研究了微生物絮凝剂的添加量、水的pH值、高岭土溶液的浓度、助凝剂的种类、助凝剂的添加量和沉淀时间对絮凝效果的影响,并研究了絮凝剂的絮凝活性分布和微生物絮凝剂的热稳定性,对絮凝效果好的菌株进行了鉴定。结果表明,菌株PY-F6的最佳絮凝条件为絮凝剂投加量30mL/L,pH 8,高岭土悬浊液浓度3g/L,Ca^(2+)的助凝效果最好,质量浓度1%的CaCl_2投加量20mL/L,静置时间20min,菌株PY-M3的最佳絮凝条件为絮凝剂投加量30mL/L,pH 8,高岭土悬浊液浓度3g/L,Ca^(2+)的助凝效果最好,质量浓度1%的CaCl_2投加量30mL/L,静置时间40min;在最佳絮凝条件下,PY-F6絮凝剂的絮凝率达到99%,PY-M3絮凝剂的絮凝率达到96%;两种微生物絮凝剂的活性成分主要存在于上清液中,为菌体细胞分泌的胞外代谢产物而非菌体本身;PY-M3絮凝剂的热稳定性较差,PY-F6絮凝剂的热稳定性较好;PY-F6菌株可以鉴定为黄三素链霉菌(Streptomyces flavotricini)。The flocculating characteristics of two microbial flocculant producing bacteria named PY M3 and PY-F6 were investigated. Main affecting factors including microbial flocculants (MBF) dosage, pH, Kaolin concentration, kind and amount of coagulant, and standing time were optimized. Moreover, the activity distribution and thermal stability of microbial flocculants were also studied, and the bacteria strain which had good flocculating effect was identified. The results showed that for the strain of PY-F6 , the optimal conditions were at MBF dosage of 30 mL/L, pH of 8, Kaolin concentration of 3g/L, using 20 mL/L of CaCl2 (mass concentration of 1%) as coagulant, standing time of 20 min. For PY-M3, the optimal conditions were at MBF dosage of 30 mL/L, pH of 8, Kaolin concentration of 3 g/L, using 30 mL/L of CaCl2 (mass concentration of 1%) as coagulant, standing time of 40 min. Under each optimized condition, the flocculating rates of PY-M6 and PY-F3 reached 99% and 96%, respectively. The activity ingredients of the two microbial flocculants were mainly extracellular metabolite which existed in the supernatant. The flocculant produced by PY-M3 had poor thermal stability whereas thermal stability of flocculant produced by PY F6 was much better. PY-F6 was identified and result showed that it was actually Streptomyces flavotricini.
