摘 要:目的:研究水飞蓟宾单用及合用肝龙含药血清对大鼠肝星状细胞增殖活性的影响及机制。方法:通过血清药理学方法制备含药血清,采用MTT法检测水飞蓟宾单用及合用肝龙含药血清对肝星状细胞增殖的影响,并用激光共聚焦测定其细胞内钙离子浓度的变化。结果:与单用水飞蓟宾相比,水飞蓟宾合用肝龙含药血清作用肝星状细胞24、48、72 h能够显著抑制肝星状细胞的增殖(P<0.01);30%水飞蓟宾联合肝龙(7.5%、15%、30%)含药血清在一定程度上可使细胞内游离Ca^(2+)浓度降低。结论:肝龙能增强水飞蓟宾在体外对肝星状细胞的抑制作用,且其作用机制可能与降低细胞内Ca^(2+)浓度有关。Objective To study the effect and mechanism of silybin single-use and combined use with ganlong capsule drug-containing serum on the proliferative activity of rat hepatic stellate cells (HSC). Methods The drug-containing serum was prepared by serum pharmacology,MTT method was used to detect the effects of silybin single-use and combined use with ganlong capsule drug-containing serum on HSC proliferation,and the changes of Ca 2+ concentration in HSC were determined via laser confocal. Results Compared with silybin single-use,silybin combined with ganlong drug-containing serum can significantly inhibit HSC proliferation at 24,48,and 72 h ( P <0.01);30% silybin combined with ganlong (7.5%,15%,30%) drug-containing serum can reduce the intracellular free Ca 2+ concentration to a certain extent. Conclusion Ganlong can enhance the inhibitory effect of silybin on HSC in vitro,and its mechanism may be related to the decrease of Ca 2+ concentration in HSC.
