摘  要:目的探讨自噬相关基因(Atg)7、Beclin-1在2型糖尿病(T2DM)患者血浆中的表达情况。方法选取无糖尿病大/微血管并发症的T2DM患者37例为T2DM组,另选取32例年龄匹配的健康人为对照组,测定各受试者血糖、血脂、糖化血红蛋白(HbA1c)、血清空腹胰岛素(FINS)等。应用稳态模型评估胰岛素抵抗指数(HOMA-IR),应用酶联免疫吸附试验(ELISA)测定血浆自噬标志物自噬相关基因(Atg)7和Beclin-1水平。结果与对照组相比,T2DM组的空腹血糖(FBG)、口服葡萄糖耐量试验中2 h静脉餐后血糖(PBG)、HbA1c、HOMA-IR、血浆Atg7和Beclin-1水平均明显升高(P<0.01,P<0.05),Atg7和Beclin-1均与高密度脂蛋白胆固醇(HDL-C)、载脂蛋白(APO)A1呈负相关(P<0.05),与FBG、PBG、HbA1c、FINS及HOMA-IR均呈正相关(P<0.05)。结论 T2DM患者血浆自噬标志物Atg7和Beclin-1的水平明显升高,自噬在2型糖尿病的病理生理进程中发挥作用。Objective To investigate the change of serum autophagy markers in type 2 diabetes patients and to analyze the relative factors. Methods 37 cases of type 2 diabetes without diabetic macrovascular complications and microvascular complications were selected as T2DM group. 32 healthy individuals were randomly selected as control. The levels of blood glucose, lipids, glycated hemoglobin and serum insulin were detected. The homeostasis model assessment of insulin resistance (HOMA-IR) was used to evaluate IR. ELISA method was applied to detect serum autophagy markers including Atg7 and Belclin-1. Results Compared with those of control group, the levels of fasting blood glucose, postprandial blood glucose, HbA1c and HOMR-IR were significantly increased in T2DM group ( P <0.01, P <0.05). Compared with those of control group, serum Atg7 and Belclin-1 levels were significantly increased in T2DM group. Both serum Atg7 and Belclin-1 were positively correlated with year, HDL, APOA1, FBG, PBG, HbA1c and HOMR-IR. Conclusions Serum Atg7 and Belclin-1 are significantly increased in T2DM patients and are positively correlated with blood glucose and HOMA-IR, indicating that autophagy plays a role in the pathophysiology and development of type 2 diabetes.
