摘  要:目的:探讨清肝益肾祛风方(QYQ)早期防治高血压及其心肌损伤的作用,为高血压的早期防治提供新思路。方法:将60只自发性高血压大鼠(SHR)随机分为模型组、清肝益肾祛风方低、中、高剂量组(QYQL, QYQM, QYQH),且以1 mg/Kg的氨氯地平(Am)、坎地沙坦(Can)分别作为阳性对照组,以同周龄WKY雄性大鼠作为正常对照组,6周龄时给予相应药物干预,共干预14周。每两周通过尾压法测量各组实验大鼠收缩压及舒张压水平,以评价QYQ的降压作用;20周龄时,取其心脏组织分别进行H&E染色及Masson's染色,以评价QYQ的心肌保护作用。结果:与正常对照组(WKY)相比,随着周龄增加,模型组(SHR)大鼠收缩压及舒张压水平均显著升高(P<0.05);与模型组相比,QYQL, QYQM, QYQH均显著降低SHR大鼠收缩压及舒张压水平(P<0.05)。与正常对照组(WKY)相比,模型组(SHR)大鼠心肌组织可见多发性灶性坏死,且具有统计学差异(P<0.05);QYQL, QYQM, QYQH均显著改善SHR大鼠心肌损伤程度,且优于Am及Can(P<0.05)。Masson's染色结果提示,与正常对照组(WKY)相比,模型组(SHR)大鼠心肌血管周围纤维化程度显著增加(P<0.05),QYQL, QYQM, QYQH均显著改善SHR大鼠心肌血管周围纤维化程度(P<0.05),各组间未见明显差异。结论:清肝益肾祛风方具有显著的降压作用,且对高血压所致心肌损伤具有显著的防治作用。Objective: To investigate the effects of QingganYishen Qufeng Fang(QYQ) on the progression of hypertension and hypertension-associated myocardial injury to provide experimental evidence supporting the clinical application of QYQ in the treatment of hypertension and hypertension-associated target organ damage such as myocardial injury. Methods: Sixty spontaneously hypertensive rats (SHRs) were randomly divided into QYQ low dosage (QYQL), QYQ medium dosage (QYQM), QYQ high dosage (QYQH) groups, positive control groups (Amlodipine treatment and Candesartan treatment) as well as vehicle-treated group. Age and sex-matched vehicle-treated WKY rats were employed as normal controls. Systolic blood pressure (SBP) and diastolic blood pressure (DBP) were measured every two weeks in the conscious rats by the tail cuff method to evaluate the effects of indicated treatments on the progression of hypertension. At the age of 20 weeks, all the rats were euthanized, followed by hematoxylin & eosin (H&E) staining and Masson’s trichrome staining to evaluate the cardioprotective effects of the indicated treatments. Results: Compared with WKY rats, SBP and DBP of SHR group were significantly increased ( P <0.05). Compared with SHRs, SBP and DBP of the QYQL, QYQM, QYQH groups as well as amlodipine and candesartan groups were significantly decreased ( P <0.05). Compared with the WKY rats, focal necrosis was readily detected in the myocardium in SHRs ( P <0.05). A better protection against myocardial injury was evident in QYQ groups rats regardless of the dosage applied compared to that from SHRs treated with amlodipine and candesartan ( P <0.05). Compared with WKY rats, perivascular fibrosis was noted in the myocardium of SHRs ( P <0.05), which was significantly attenuated as a result of QYQL, QYQM, and QYQH groups( P <0.05). No significant differences were observed among the QYQL, QYQM, QYQH groups. Conclusion: QYQ can significantly attenuate the progression of hypertension in SHRs, meanwhile, conferre a better protection against the
