摘  要:目的:探讨电针法对围绝经期大鼠的神经保护作用及其机制研究。方法:利用双侧卵巢切除(OVX)的方法制备围绝经期大鼠模型,SD大鼠随机分为假手术组、模型组、雌激素组和电针组。采用原位末端标记法(TUNEL)检测海马区细胞凋亡、尼氏染色检测神经元结构、免疫荧光染色法检测海马区NF-M表达情况、Western Blot法检测脑源性神经营养因子(BDNF)、环磷腺苷效应元件结合蛋白(CREB)、p-CREB蛋白表达情况。结果:与假手术组相比,模型组大鼠海马区神经细胞凋亡数量增加,神经元排列疏松散乱(P <0. 01),NF-M荧光强度减弱(P <0. 01),BNDF蛋白表达减少、p-CREB/CREB比值降低(P <0. 01)。经电针和雌激素治疗后,大鼠海马凋亡细胞数量减少,神经元排列整齐数量增加,NF-M荧光强度增加,BNDF蛋白表达增加、p-CREB/CREB比值增加(P <0. 01)。结论:电针对围绝经期大鼠具有一定的神经保护作用,可能与上调脑组织CREB-BDNF信号通路主要蛋白表达有关。Objective: To investigate the neuroprotective effects of electroacupuncture and possible mechanism in the perimenopausal rats. Methods: SD rats were randomly divide into the sham group,the model group,the estrogen group and the electroacupuncture group. In this study,the perimenopausal model in rats was established by ovariectomy. TUNEL was used to detect apoptotic cells expression and Nissl staining detected the hippocampus neuron structure,immunohistochemistry staining was used to detect NF-M expression,and Western Blot was used to detect BDNF,CREB and p-CREB protein expressions. Results: Compared with the sham group,the number of apoptotic cells were increased in the model rats’ hippocampus and the neurons were scattered. In addition,NF-M fluorescence intensity was decreased and the expressions of BNDF protein and p-CREB/CREB ratio were decreased. Furthermore,both the electroacupuncture group and the estrogen group’s number of apoptotic cells were significantly decreased. The structure of hippocampus neurons was normal. NF-M fluorescence intensity was increased and the expression of BNDF protein and p-CREB/CREB ratio was increased. Conclusion: The electroacupuncture has neuroprotective effect on perimenopaual rats,and may be related to the up-regulated proteins expression of CREB-BDNF signaling pathway.
