摘  要:目的:观察电针双侧"足三里"穴对脾气虚模型大鼠肌肉组织内线粒体自噬相关因子MUL1和ULK1基因及蛋白表达的影响。方法:32只SD雄性大鼠按随机数字表法分为4组:空白组、脾气虚组、治疗组、非经非穴组,每组8只。采用劳倦过度和不规则饮食复合法复制脾气虚证大鼠模型,依据造模评定标准造模成功后,治疗组大鼠给予电针双侧"足三里"穴治疗处理,非经非穴组大鼠给予电针双侧非经非穴点干预处理,1次/d,20min/次,空白组及脾气虚组大鼠在操作台上固定20min,不给予其它处理。干预7d后取胃及骨骼肌组织,采用荧光定量PCR法检测大鼠胃及骨骼肌组织MUL1和ULK1 mRNA表达水平,蛋白质免疫印迹法(WB)检测大鼠胃及骨骼肌组织内MUL1和ULK1蛋白含量变化。结果:脾气虚组和非经非穴组大鼠胃及骨骼肌组织内MUL1 mRNA和蛋白表达水平均明显高于空白组,ULK1 mRNA及蛋白表达下降(P <0. 05);治疗组大鼠胃及骨骼肌组织MUL1 mRNA和蛋白表达水平相比于脾气虚组有所下降,ULK1mRNA及蛋白表达升高(P <0. 05);非经非穴组同脾气虚组相比无统计学意义(P <0. 05)。结论:电针"足三里"穴可以从转录水平上抑制脾气虚大鼠肌肉组织内MUL1的过表达,稳定ULK1的调节作用,参与线粒体自噬的调控作用进而改善脾气虚证。Objective: To observe the effect of electroacupuncture at bilateral Zusanli( ST36) on the expressions of mitochondrial autophagy related factors MUL1 and ULK1 gene and protein in muscle tissue of rats with spleen Qi deficiency model. Methods:Thirty-two male SD rats were randomly divided into 4 groups: blank group,spleen Qi group,treatment group and non-acupoint group( n = 8). The model of spleen Qi deficiency syndrome was established by the composite methods of improper diet and overstrain. When the model was successfully established according to the model evaluation standard,the rats were treated with electroacupuncture at the bilateral Zusanli( ST36) in the treatment group,and the non-acupoint group were treated with electroacupuncture at bilateral non-acupoint. The blank group and spleen deficiency group were fixed on the operating table with no other treatment,1 time/day,20 min/time. The stomach and skeletal muscle tissue of rats were taken out after seven days intervention.The changes of MUL1 and ULK1 mRNA and protein expression in skeletal muscle and gastric smooth muscle were detected by fluorescence quantitative PCR method and Western blot( WB). Results: The protein and mRNA expression of MUL1 in skeletal muscle and gastric smooth muscle of were significantly higher than those of the blank group,and ULK1’s mRNA and protein expression in skeletal muscle and gastric smooth muscle were significantly lower than those of the blank group,spleen Qi deficiency group and non-acupoint group. Compared with the group of spleen Qi deficiency,the level of MUL1 decreased and the level of ULK1 increased in treatment group and there was no significant difference between spleen Qi deficiency group and non-acupoint group( P < 0. 05). Conclusion: Electroacupuncture at Zusanli( ST36) can participate in the regulation of mitochondrial autophagy which stabilize the regulation of ULK1 by inhibiting the over expression of MUL1 in muscle tissue of rats with spleen Qi deficiency at the transcriptional level,thereby improving sple
