摘 要:目的:探讨全真一气汤增强慢性阻塞性肺病(Chronic Obstructive Pulmonary Disease,COPD)大鼠气道抗微生物免疫功能机制。方法:建立COPD大鼠模型,以中药全真一气汤干预后,实时荧光定量聚合酶链反应(PCR)检测肺组织线粒体超氧化物歧化酶-2(SOX2)表达水平的变化,Western blot检测肺组织中SOX2蛋白水平的变化。结果:中药组较模型组SOX2mRNA表达增强,其表达丰度分别为模型组的1.83倍;中药组较模型组SOX2蛋白表达上调。结论:提示COPD大鼠肺组织SOX2表达下降,这可能是COPD患者气道易感的重要机制,全真一气汤能通过增强SOX2的表达及活化,从而提高COPD大鼠气道对病原体的免疫能力。Objective:To investigate the mechanism of the Quanzhen Yiqi decoction (全真一气汤) in enhancing airway antimicrobial immunity of COPD rats.Methods:A rat model of COPD was established.After the intervention of the Quanzhen Yiqi decoction,the expression of SOX2 in lung tissue was detected by real-time fluorescent quantitative PCR.The change of SOX2 protein level in lung tissue was detected by Western blot.Results:Compared with the model group,the expression of SOX2 mRNA was increased in the TCM medicine group;the expression abundance was 1.83 times of the model group.The expression of SOX2 protein was up-regulated in the TCM medicine group.Conclusion:The expression of SOX2 in lung tissue of COPD rats is decreased,which may be an important mechanism of airway susceptibility in COPD patients.The Quanzhen Yiqi decoction can enhance the immunity of COPD rats,to airways by enhancing the expression and activation of SOX2.
