摘  要:目的:研究苦荞碱性提取物(FTAE)对2型糖尿病大鼠的降糖作用以及对α-葡萄糖苷酶的抑制作用。方法:通过高糖高脂喂养大鼠4周后腹腔注射链脲佐菌素(STZ,35 mg·kg^(-1))复制大鼠2型糖尿病模型。将成模大鼠随机均分为模型组,阿卡波糖组(32 mg·kg^(-1)),FTAE高(86 mg·kg^(-1))、低剂量(43 mg·kg^(-1))组,以健康正常大鼠作为空白对照组。灌胃给药,bid。连续给药4周后,检测大鼠空腹血糖(FPG)值,计算大鼠时间-血糖曲线下面积(AUC)。连续给药6周后,检测大鼠血清胰岛素(INS)与糖化血红蛋白(Hb Alc)水平,计算胰岛素抵抗指数(HOMA-IR)。考察FTAE对α-葡萄糖苷酶的抑制作用。结果:与模型组比较,FTAE组在给药后第6周FPG显著降低(P <0. 05或P <0. 01),FTAE高、低剂量组分别在给药后第6周和第2周开始FPG与阿卡波糖组差异有统计学意义(P <0. 05或P <0. 01)。与模型组比较,FTAE组大鼠AUC显著降低(P <0. 05或P <0. 01),FTAE低剂量组与阿卡波糖组比较差异有统计学意义(P <0. 01)。与模型组比较,FTAE高剂量组的INS、HOMA-IR、Hb Alc水平显著降低(P <0. 05或P <0. 01),FTAE低剂量组HOMA-IR水平显著降低(P <0. 05),FTAE各组与阿卡波糖组比较差异有统计学意义(P <0. 05或P <0. 01)。随着FTAE浓度的增加,对α-葡萄糖苷酶抑制率逐渐增加,半数抑制浓度为(116. 468±1. 065) mg·L^(-1),与阿卡波糖组比较差异有统计学意义(P <0. 01)。结论:FTAE对高糖高脂联合STZ诱导的2型糖尿病模型大鼠有降糖作用,其机制可能与其抑制α-葡萄糖苷酶活性有关。Objective:To study the hypoglycemic effect of alkaline extracts from Fagopyrumtataricum(L.)Gaertn(FTAE)in type 2 diabetic rats and its inhibitory effect onα-glucosidase.Methods:After fed with high sugar and fat for 4 weeks,the rat model of type 2 diabetes mellitus was induced by intraperitoneal injection of streptozotocin(STZ,35 mg·kg^-1).The type 2 diabetic rats were randomly divided into the model group,acarbose group(32 mg·kg^-1),FTAE high(86 mg·kg^-1)and low(43 mg·kg^-1)dose groups,and the healthy normal rats were used as the blank control group.The drug was administered by gavage,twice a day.After 4 weeks of continuous administration,the fasting blood glucose(FPG)was measured and the area under time-blood glucose curve(AUC)was calculated.After 6 weeks of continuous administration,the serum insulin(INS)and glycosylated hemoglobin(Hb Alc)levels were measured and insulin resistance index(HOMA-IR)was calculated.The inhibitory effect of FTAE onα-glucosidase was investigated.Results:Compared with that in the model group,FPG in FTAE groups decreased significantly in the 6 th week after the administration(P<0.05 or P<0.01),and there was significant difference between FTAE groups and acarbose group in the 6 th and 2 nd week after the administration(P<0.05 or P<0.01).Compared with that in the model group,AUC in FTAE groups was significantly lower(P<0.05 or P<0.01),and there was significant difference between FTAE low dose group and acarbose group(P<0.01).Compared with those in the model group,the levels of INS,HOMA-IR and Hb Alc in FTAE high dose group were significantly lower(P<0.05 or P<0.01),and the level of HOMA-IR in FTAE low dose group was significantly lower(P<0.05).There were significant differences between FTAE groups and acarbose group(P<0.05 or P<0.01).With the increase of FTAE concentration,the inhibition rate ofα-glucosidase gradually increased,and the half inhibition concentration was(116.468+1.065)mg·L^-1,which had significant difference from acarbose(P<0.01).Conclusion:FTAE has hypoglycemic eff
