摘 要:为了鉴定分离自甘肃永靖的1株双芽巴贝斯虫Hsp20基因型为一新基因型,参考双芽巴贝斯虫Hsp20核苷酸序列,设计特异性引物。对其进行扩增,利用生物信息学方法对其进行比对分析。结果显示,扩增获得了700 bp的核苷酸片段。与GenBank中公布的18种不同物种的氨基酸序列构建遗传发育树,结果表明本实验获得的双芽巴贝斯虫与美国株和中国新疆株的亲缘关系最近。对其氨基酸序列进行比对分析,发现在第31~85位点存在冗余,与公布的Hsp20氨基酸序列存在明显的差异。通过同源建模分析,发现该冗余致使Hsp20蛋白在第7号位形成1个环形结构,从而引起了第3号位的拉伸。通过生物信息学分析得出,获得的双芽巴贝斯虫甘肃株的Hsp20基因是一新基因型。该基因型的存在为更加准确诊断双芽巴贝斯虫病提供了依据。In order to detect Hsp20 of Babesia bigemina isolated from Yongjing,Gansu,the specific primers were designed with reference to the Hsp20 nucleotide sequence of B.bigemina.The sequence was amplified and compared using bioinformatics methods.In result,a nucleotide fragment of 700 bp was obtained by amplification and was compared with 18 other amino acid sequences in Gen Bank for a phylogenetic tree.The results showed that the B.bigemina identified in this experiment had closest relationship with the US and Xinjiang strains.The amino acid sequence was aligned and analyzed,and there was redundancy at the 31-85 position,which was significantly different from the previously published Hsp20 amino acid sequences.Homology modeling analysis showed this redundancy caused the Hsp20 protein to form a ring structure at the 7 th position,which caused stretching at the 3 rd position.According to the bioinformatics analysis,the Hsp20 gene of the B.bigemina strain was a new genotype.The presence of this genotype provided a basis for a more accurate and reliable diagnosis of the disease caused by B.bigemina.
