摘 要:通过2,4-二羟基苯甲醛和对氨基水杨酸(PAS)反应合成了新型席夫碱及镍配合物。利用~1H核磁共振、质谱、红外光谱、元素分析等方法对其结构进行表征。采用荧光光谱法研究了配合物和牛血清白蛋白(BSA)的相互作用。结果表明,配合物对BSA猝灭机制为静态猝灭。计算了不同温度下相互作用的结合常数、结合位点数及热力学参数,推断二者作用力的类型为氢键和范德华力。抑菌实验结果表明,配合物的抑菌能力明显大于中心原子和配体组,对金黄色葡萄球菌的抑菌能力大于大肠杆菌,镍配合物对大肠杆菌和金黄色葡萄球菌的最低抑菌浓度分别为5.00μg·mL^(-1)和2.50μg·mL^(-1)。2,4 Dihydroxybenzaldehyde-4-aminosalicylic acid Schiff base was synthesized from 2,4 dihydroxybenzaldehyde and 4-aminosalicylic acid(PAS )in methanol solution,and its nickel complex was obtained by the reaction of the Schiff base ligand and nickel ion 1H NMR,mass spectrometry,FT IR,elemental analysis,etc were used to characterize the structure of the ligand and its nickel complex. The interaction between the nickel complex and bovine serum albumin(BSA )were studied by fluorescence spectros copy. Experimental results indicated that the quenching mechanism was static quenching. The binding constants,the number of binding sites and the thermodynamic parameters of the BSA/ complex system were calculated at different temperatures,indicating their interaction forces were mainly hydrogen bonds and van der Waals forces. In addition,the antibacterial results showed that the antibacterial activities of the nickel complex were significantly greater than that of the ligand and the nickel ion. And the antibacteri al activities against Staphylococcus aureus were greater than that of E. coli. The minimum inhibitory concentrations of the nickel complex against E. coli and Staphylococcus aureus were 5 00 μg mL^-1 and 2 50 μg mL^-1,respectively .
