摘 要:目的探讨姜黄素通过抑制Janus激酶2-信号转导转录激活因子3(JAK2/STAT3)信号通路改善脂多糖诱导的小鼠急性肺损伤的机制。方法将30只雄性小鼠分为对照组、模型组和姜黄素干预组。姜黄素干预组连续灌胃给予姜黄素(100 mg/kg)7 d,每日1次,末次给药1 h后模型组和姜黄素腹腔注射给予脂多糖(10 mg/kg),对照组给予腹腔注射等量生理盐水,6 h后麻醉处死大鼠,进行小鼠肺组织切片观察,计算大鼠肺组织湿干重(W/D)比,检测支气管肺泡灌洗液中蛋白、肿瘤坏死因子α(TNF-α)和白介素-8(IL-8)水平,同时检测肺组织中JAK2、p-JAK2、STAT3和p-STAT3蛋白表达水平。结果对照组小鼠肺组织切片未见明显异常,模型组小鼠肺组织切片见肺泡壁部分增厚,细支气管腔内有大量脱落炎细胞及渗出物,泡壁毛细血管充血严重,姜黄素干预组小鼠肺组织病理症状较模型组轻;与对照组比较,模型组肺组织W/D和支气管肺泡灌洗液中蛋白、TNF-α、IL-8含量增加,给予姜黄素干预后,上述指标均降低;小鼠肺组织中JAK2和STAT3蛋白表达变化不显著,模型组小鼠肺组织中p-JAK2和p-STAT3蛋白表达较对照组增加,给予姜黄素干预后,小鼠肺组织中p-JAK2和p-STAT3蛋白表达降低。结论姜黄素可以通过抑制JAK2/STAT3信号通路减轻脂多糖诱导的小鼠急性肺损伤。Objective: To investigate the effect of curcumin on LPS-induced acute lung injury in mice based on JAK2/STAT3 signaling pathway. Methods: 30 male mice were divided into the control group,the model group and curcumin intervention group. Rats were made into LPS-induced acute lung injury models,except those in the control group. Then the lung tissue sections of mice were observed,the W/D of lung tissue in rats was calculated,and the levels of protein,TNF-α and IL-8 in bronchoalveolar lavage fluid were measured. At the same time,the expression levels of JAK2,p-JAK2,STAT3 and p-STAT3 proteins in lung tissues were detected. Results: There was no obvious abnormality in the control group. The alveolar wall of the model group was partly thickened. There were a large number of exfoliated inflammatory cells and exudates in the bronchioles. The alveolar capillary congestion was serious. The pathological symptoms of lung tissue in curcumin intervention group were milder than those in model group. Compared with the control group,the W/D of lung tissue and the contents of protein,TNF-alpha and IL-8 in bronchoalveolar lavage fluid increased in the model group. After curcumin intervention,the above indexes decreased. The expression of JAK2 and STAT3 in lung tissue of mice was not significantly changed. The expression of p-JAK2 and p-STAT3 in lung tissue of model group was higher than that of control group. After curcumin intervention,the expression of p-JAK2 and p-STAT3 in lung tissue of mice decreased. Conclusion: Curcumin can alleviate LPS-induced acute lung injury in mice by inhibiting JAK2/STAT3 signaling pathway.
